Objective:In this study,we observed the changes of neurotransmitters and aquaporins in lung tissue of rats with constipation,analyzed the pathological process of intestinal dryness and damage of lung yin,so as to clarify the biological basis of enteropathy and lung,and used the famous prescription "Tongbian Decoction" to intervene in the animal model.At final,from the point of view of modern medicine,to explain the scientific nature of traditional Chinese medicine for the treatment of intestinal disease from lung.Methods:60 SD rats were randomly divided into nonnal group(n = 20)and model group(n =40),The rats were weighed for 120 days,and the morphology,quantity and water content of the rats were observed.Randomly selected normal group and model group with 10 rats in each observation.To observe the intestinal transmission function,and the changes of neurotransmitter SP,VIP and aquaporin AQP1 and AQP3 in lung tissue of constipation model rats were observed by ELISA and immunohistochemistry.The 30 rats were randomly divided into model group,decoction group and polyethylene glycol powder group.Each group had a total of 10 rats.The rats were treated with drug intervention in January.Record and detect the above datas.Result:1.The rats in the model group,the number of defecation stool water content and rat intestinal and colonic fecal retention rate of carbon propelling were significantly lower than normal group(P<0.01).After the modeling,the content of SP is significantly lower than the normal group model group(P<0.01)in the lung,intestinal tissue in SP model group decreased compared with the normal group(P<0.05);the content of VIP group with lung model significantly increased compared with normal group(P<0.01),VIP content in colon tissue compared with normal group increased(P<0.05).At the end of the model,the amount of AQP3 and AQP1 in the lung tissue of model group was higher than that of normal group(P<0.01).2.After 4 weeks of drug intervention,rats in the model group,the number of defecation and fecal water content of rat intestinal and colonic fecal retention rate of carbon propelling than in the normal group were significantly reduced(P<0.01),Tongbian Decoction and Forlax group rat feces number,stool moisture and fecal retention colon were significantly improved compared with the model group(P<0.01),To promote the rate of Tongbian Decoction soup group compared with model results there are significant differences in the rat small intestine carbon black(P<0.01),there was statistical significance comparing Forlax group with model group(P<0.05);the Tongbian Decoction in improving the number of rats defecation than group B(P<0.01),and Tongbian Decoction in improving the large fecal water content,rat small intestine carbon ink advance rate were better than group B(P<0.05),and the two group in improving the colon of rats had no significant difference between the left hand stool(P>0.05).3.After 4 weeks of drug intervention,the content of SP and VIP in lung tissue of model group was significantly lower than that of normal group(P<0.01);Laxative soup and Forlax intervention compared with model group,the content of SP in lung tissue were increased,The content of SP in lung tissue of Tongbian Decoction group changed obviously(P<0.01),The two groups after the intervention,the content of SP in colon of rats was significantly higher than that in model group(P<0.05);after the treatment of Forlax group and Tongbian Decoction group rat pulmonary intestinal VIP content compared with the model group were significantly reduced(P<0.01),Laxative soup and Forlax after the intervention,the content change of VIP two group SP,lung and intestine had no statistical significance(P>0.05)4.The results of immunohistochemistry showed that AQP1 and AQP3 in lung and intestine in the model group IOD values were higher than the normal group(P<0.01),and the dyeing range and depth were increased compared to controls,laxative soup and Forlax after the intervention of aquaporin content in lung and colon tissue were lower than those in the model group(P<0.01),Tongbian Decoction group compared with Forlax group decreased significantly(P<0.01).The expression of lung AQP1 and AQP3 related protein Western blot detection after drug intervention,the results showed that the expression of AQP3 protein and AQP1 in colon tissue of lung tissues in model group were significantly higher than those of normal group(P<0.01),laxative soup and Forlax intervention,compared with the model group were decreased AQP1 and AQP3 expression in lung and intestine.The statistical results were statistically significant(P<0.05).Conclusion:1.A new type of chronic transit constipation model was successfully developed by using compound Acetophenone.2.The SP,VIP and AQP1 of intestinal tract,intestinal tract and AQP3 may be the biological basis of constipation and intestinal disease.3.Tongbian Decoction can significantly improve the symptoms of constipation in rats,promote intestinal peristalsis,and reduce the amount of retained stool in the colon4.Tongbian Decoction for constipation may be possible to increase the level of SP in the lung tissue,lower the content of VIP in the lung and the content of AQP1 and AQP3 protein in the intestine,so as to achieve the effect of constipation.
