Due to the limitation of the current mainstream medical imaging technology, clinical diagnosis can rely on the data of two-dimensional tomographic image only. Doctors speculate the spatial location of lesion and the mutual relationship between lesions and human tissues by two-dimensional tomographic image data.However, with a great subjectivity in such diagnosis, clinical diagnostic realm requires an effective tool so as to observe human organs and tissues comprehensively. Then medical image three-dimensional reconstruction system came into being.Firstly, the paper describes DICOM standard of two-dimensional tomographic image data file, analyzed the role and significance on files structure partial in DICOM standard emphatically, and DICOM file instances were analyzed. Introduced the basic process of three-dimensional image reconstruction, mainly analyzed the types and characteristics of image data on three-dimensional reconstruction.Secondly, the paper studies two types of three-dimensional image reconstruction algorithms. For surface rendering algorithm, analyze two important concepts in the reconstruction process for voxel model and isosurface; For volume rendering algorithm,analyzed technical details of voxel classification and color transparency assignments in volume rendering process.The paper not only studies Marching Cube and Dividing Cube in surface rendering algorithm, but also Ray Casting, Texture Mapper 3D and MIP Ray Casting in volume rendering algorithm. Pros and cons had been made in rendering speed and display effect of various algorithms as a way to analyze clinical applications according to the characteristics of different algorithms.Finally, the paper based on VTK, in accordance with object-oriented design idea, taking advantage of MFC interface design techniques, implemented based on VTK medical image three-dimensional reconstruction system by cross-platform project build tool-CMake. Through a reconstruction from a sequence of two-dimensional tomographic image, it demonstrates multiple functions like reconstruction, translation, rotation and interception, its design requirements and expectations had been accomplished.
