Like human's blood vessel, Plant's vascular bundle transfers nutrition and water to all its organizations. With three-dimensional Reconstructing technology, we can simulate the vascular bundle channel, compute the section area, and make people to observe or measure the plant in detail. It is very helpful for people to find out plant's inner structure. This paper mainly discussed three methods of 3D Reconstruction technology, include 3D Reconstruction based on contour extraction, Surface Rendering based on voxels, and Volume Rendering based on voxels.The paper discussed and solved the following problems. Firstly, realize the image registration with affine transformation. Secondly, realize the extraction and reservation of character points on contours with methods of multi-contour tracing and difference code representing, meanwhile, 3D Reconstruction adopt the method of linking the according character points on neighboring slice contour to construct triangle meshes.Thirdly, research the Marching Cubes algorithms to extract the iso-surface about Surface Rendering .At last ,we discussed the Volume Rendering and some algorithms like Ray-Casting, Splatting, Shear-Warp etc. Also with the help of VTK, we realized the Volume Rendering with the slice images of cucumber.To obtain better results of 3D Reconstruction, the paper also discussed image processing algorithms,Light,Texture,depth-buffer in OpenGL and VTK. At last, we complete the 3D Reconstruction of image slices.
