Background: As one of the most common malignant digestive tumor,colorectal cancer has become a great threat to the health of human.The pathogenesis of colorectal cancer is various and complex,which is mainly regulated by a serious of abnormally-expressed oncogenes and tumor suppressor genes.The key factors and signal pathways underlying this are still unclear now.To study it would be of great social and economic value.EAG1,HIF-1?,and VEGF were typical oncogenes and have been confirmed playing important roles in tumor occurence and development.A positive correlation among them has also been confirmed in many other cancers,but the expression and function of EAG1,HIF-1?,and VEGF in colorectal cancer has not been reported yet.Purpose: This article is trying to find out the expression of EAG1,HIF-1?,VEGF in colorectal cancer and colorectal adenomas,their correlationship and the relation of them with biological behaviours of colorectal cancer.Methods: The tested samples of colorectal cancer(n=73),colorectal adenomas(n=26)and healthy tissues(n=10)were collected from pathological department of MaWangDui hospital of Hunan province.Real-time Quantitative PCR and IHC assays were taken to study the mRNA and protein levels of EAG1,HIF-1?,VEGF in colorectal cancer,colorectal adenomas and healthy tissues.The statistic analysis was used to analyse the correlation of these three genes.Results: The results of Q-PCR showed that the mRNA levels of EAG1 in colorectal adenomas and colorectal cancer tissues were 1.6 times and 4.5 times higher than that in normal colorectal specimens,respectively(P<0.05);mRNA levels of HIF-1? in colorectal adenomas and colorectal cancer tissues were 1.2 times and 2.2 times higher than that in normal colorectal specimens,respectively(P<0.05);mRNA levels of VEGF in colorectal adenomas and colorectal cancer tissues were 1.61 times and 3.14 times higher than that in normal colorectal specimens,respectively(P<0.05).The IHC test data show that there were significant difference of protein expriession among three groups.In colorectal cancer tissues,the positive rate of EAG1,HIF-1?,VEGF were 86.30%,93.15%,89.04%,respectively,much higher than that in colorectal adenomas and normal tissues(P<0.05).Further,EAG1,HIF-1?,VEGF expression levels showed a strong correlation with Dukes classification,differentiated degree,tumor infiltration and metastasis levels of colorectal cancer(P<0.05).The statistic analysis of the IHC data also showed that the expression of EAG1,HIF-1?,and VEGF were significantly related with each other(Rs>0.5,P<0.05).Conclusion: EAG1,HIF-1?,VEGF were significantly up-regulated in colorectal cancer tissues,and there were positively correlations among the three genes;the expression of EAG1,HIF-1?,VEGF with significant correlated with Dukes classification,differentiated degree of tumor,infiltration and transfer situation,but not with the age and sex of patients.
