Objectives: Colorectal carcinoma is one of the common malignantgastrointestinal tumors.The destruction of balance mechanism between theinactivation of the cancer suppressor genes and activation of cancer genes isone of the main factors.Gene NGX6,who is a tumor suppressor gene, candecelerate or inhibit the blossom of tumor,by means of gearing downproliferation of the cell cycle,regulating expression of cycle protein in cell,and inhibiting angiogenesis of tumor.As a tumor suppressor gene,IL-17F iscapable to subdue the formation of colorectal cancer,through decreasingVEGF and inhibiting angiogenesis of tumor.VEGF is an important angiogenicfactors,who plays a key role in the development of cancer.At present,theeffect of IL-17F on the colorectal cancer has been seldom reported indomestic and abroad. Investigation on combined test for the influence ofNGX6and IL-17F on colorectal cancer and adenomas has not been reportedbefore.The experiment has studied the expression of NGX6and IL-17F incolorectal cancer and adenoma, investigated the correlation between VEGFand them,analyzed the relationship among the three and data of clinicalpathological.Methods:Step one:Selecting60cases with pathological diagnosis as colorectalcancer or adenoma form gastrointestinal surgical excision specimens,whichwere sealed by wax,in the People’s Hospital of Sichuan Provincial.Step two:Extract96cases of adenomas, among which there will be each24cases for a villous adenoma, tubular adenoma, tubular-a villous adenoma, and serrated adenoma,according to the degree of hyperplasia, which weredivided into mild, moderate,severe,8cases for each. in addition,8cases ofnormal tissues of left half colon form the healthy check-up crowd wereselected as control group.Step three:Envision-two-step of immunohistochemistry is used to test theexpressions of NGX6and IL-17F in colorectal cancer and colorectaladenomas.The experiment would analyze what is the correlation betweenVEGF and them, and investigate the correlation among the clinicalpathological data and the three at the same time.Statistical Methods: The analysis of data is performed by statisticalsoftware SPSS13.0, with α=0.05as the inspection level.Results:1.The positive expression rate of NGX6in colorectal cancer, adenomasand normal tissue is41.67%,65.63%,100%respectively, among which thedifference was statistically significant (P<0.05);there are significantdifferences with P <0.01between colorectal cancer and adenoma, so as tocolorectal cancer and normal group;difference between adenoma and normaltissue was statistically significant (P <0.05).2.The expression of NGX6in colorectal cancer is associated withinfiltration degree of serosal and the presence of lymph node metastases.Thepositive expression rate is24.3%and69.6%respectively in infiltration serouslayer and no infiltration serous layer,the difference between them is obviouslysignificant (P<0.01).Meanwhile,the positive expression rate is17.4%and56.8%respectively in lymph node metastasis group and no lymph nodemetastasis group,the difference among them is obviously significant(P<0.01).The difference them is obviously significant.There are significant differences(P<0.01) among the group of stage I+II and the group of III+IVin TNM stage with the positive expression rate59.38%and21.43%respectively.The expression of NGX6in colorectal cancer has nothing to dowith patients’ age,sex,the lesion location and differentiated degree(P>0.05).3.The positive expression rate of IL-17F in colorectal cancer,adenomas and normal tissue is56.7%,70.83%,100%respectively, amongwhich the difference was statistically significant (P <0.05).Differencebetween colorectal cancer and normal tissue was statistically significant (P <0.05).There are not statistical significance with P>0.05between colorectalcancer and adenoma, so as to adenoma and normal group.4.The expression and of IL-17F in colorectal cancer is associated withdegree of differentiation, infiltration degree of serosal and the presence oflymph node metastases.The positive expression rate is75%,50%respectivelybetween group of high differentiation, and group of differentiation of low andmiddle, the difference was statistically significant (P <0.05).Differences arestatistically significant(P<0.05)between no infiltration serous layer andinfiltration serous layer with the positive expression rate of78.26%and48.65%respectively.The positive expression rate is83.78%and26.09%respectively in no lymph node metastasis group and lymph node metastasisgroup, the difference among them is obviously significant (P<0.01).Whilethere was no statistically significant difference (P>0.05) among the patients’age,sex,the lesion location and TNM stage during the expression of IL-17F incolorectal cancer.5.The positive expression rate of VEGF in colorectal cancer, adenomasand normal tissue is41.67%,65.63%,0%respectively, among which there aresignificant differences with P <0.01;differences of comparing one to another are statistically significant (P <0.05).6.The expression of IL-17F and NGX6in colorectal cancer arepositively correlated, with0.276as the correlation coefficient (P <0.05).Theexpression of NGX6and IL-17F in colorectal cancer are respectivelynegatively correlated with the expression of VEGF in colorectal cancer, andtheir correlation coefficient are respectively-0.348,-0.274(P <0.01).7.The positive expression rate of NGX6in villous adenoma, tubular-avillous adenoma,tubular adenoma and serrated adenoma are54.12%、58.33%、70.83%、79.17%respectively; the positive expression rate of IL-17Fin which are58.33%、66.67%、79.17%、79.17%respectively; the positiveexpression rate of VEGF in which are62.50%、45.83%、41.67%、33.33%respectively; differences among the four kinds of adenoma for therespectively three factors have no statistical significance (P>0.05).Differences are statistically significant(P<0.05)among mild, moderate,severe,which were divided according to the different degree of hyperplasia inthe four kinds of adenoma.Conclusions:1.The expression of NGX6and IL-17F in colorectal cancer decreasedobviously or absent, while in normal colon tissue they are strongly expressed.The expression of NGX6and IL-17F in colorectal cancer are cut down in allsorts of mild-to-moderate hyperplasia of adenoma, which decreased obviouslyor absent in adenoma of severe hyperplasia or adenoma canceration.While theexpression of VEGF in adenoma rises, predominantly for the adenoma ofsevere hyperplasia.These prompted that NGX6,IL-17F and VEGF areinvolved in the process from adenoma to cancelation.2.The expression of NGX6in colorectal cancer is associated with infiltration degree of serosal, the presence of lymph node metastases andTNM stage.The expression of NGX6in colorectal cancer cut obviously ormissing,when there is infiltration serous layer;or there is lymph nodemetastasis; or it is belong to the stage of middle or late in TNM stage, whichprompt that the expression of NGX6may be associated with prognosis ofcolorectal cancer.3.The expression of IL-17F in colorectal cancer is associated withinfiltration degree of serosal, the presence of lymph node metastases anddifferentiated degree.The expression of IL-17F in colorectal cancer cutobviously or missing, when there is deep infiltration of serous layer;or there islymph node metastasis;or it is belong to poorly differentiated carcinoma oradenocarcinoma,which prompt that the expression of IL-17F may beassociated with the development of colorectal cancer,and be helpful toevaluate prognosis.4.The expression rate of NGX6and IL-17F in colorectal cancer ispositive correlated with each other.The expression rate of NGX6and IL-17Fin colorectal cancer is negative correlated with the expression rate of VEGFrespectively,which prompt that the NGX6and IL-17F,as tumor suppressorgene, inhibit the occurrence of tumors,by means of reducing the expression ofVEGF and inhibiting angiogenesis of tumor.
