Objective:To research the K-ras protein and HDAC6 protein in colorectal carcinoma,colorectal adenoma and normal colorectal mucosa tissues,and to explore the relationship between K-ras and HDAC6 in occurrence and development of colorectal cancer and colorectal adenomas.And to provide a theoretical basis for pathogenesis and early detection of colorectal cancer.Methods : 1.Collecting the 64 specimens removed by colonoscope and pathologically diagnosed as colorectal cancer tissues,normal colorectal mucosa tissues and colorectal adenomas tissues in the Operating room of Digestive Endoscopy of The Affiliated Hospital of Southwest Medical University during April,2015 to July,2016.The normal colorectal mucosa group isthat adjacent normal tissue more than 5cm from the tumor tissue with colorectal cancer patients.Colorectal cancertissues and colorectal adenomas tissues are classified as experimental group,normal colorectal mucosa tissues specimens as control group.K-ras and HDAC6 protein are detected by immunohistochemical methods.2.It is collected the relevant clinical andpathological data statistics of selected cases including patients' gender,age,tumor size,and the differentiation degree and location of pathologic histology.The SPSS 20.0 edition was used for data analysis.The Chi-square testwas used for comparison of data between the two groups.The correlation of categorical variables is described by Spearman correlation coefficient.P-value < 0.05 wasconsidered statistically significant.Results:1.The expression rate of K-ras protein in normal colorectal mucosa,colorectal adenomas and colorectal cancer tissue group was 11%(7/64),(43/64),76%(49/64),respectively.Theexpression rate between colorectal adenomas tissue group and colorectal cancer tissue group was not statistically significant(P > 0.05),while the expression rate between normal colorectal mucosa group and colorectal cancer group was statistically significant(P < 0.05),and the expression rate between normal colorectal mucosa and colorectal adenoma group was statistically significant(P < 0.05).2.The expression of HDAC6 proteinwas successively increasing in normal colorectal mucosa,colorectal adenomas and colorectal cancer group,whose expression rate was 7%(5/64),(18%),and 41/64(12/64),respectively.The expression rate between HDAC6 protein in colorectal adenoma and colorectal cancer tissue groupwas statistically significant(P < 0.05),and the expression rate between normal colorectal mucosa and colorectal cancer group wasstatistically significant(P < 0.05),while the expression rate between normal colorectal mucosa vand colorectal adenoma group was not statistically significant(P >0.05);3.The expression level of HDAC6 protein in colorectal cancer tissue group on patients' gender,age,tumor sizeand tumor position was not statistically significant(P > 0.05),while it was statistical significance on histological differentiation degree of colorectal cancer tissue(?2=6.208,P =0.045).The lower the degree of differentiation of colorectal cancer tissuewas,the higherthe positive expression rate of HDAC6 protein became;4.The correlation of K-ras protein and HDAC6 protein was analyzedin the colorectal adenomastissue and colorectal cancer tissue group expression by Spearman.The result showed that there was no significant correlation between the expression of K-ras protein and HDAC6 protein in colorectal adenoma group(P ? 0.05),while there was a positive correlation between the expression of K-ras protein and HDAC6 protein in colorectal cancer tissue group(P<0.05).The spearman correlation coefficientwas0.277.Conclusions:1.The tendency of increasing expression rate of K-ras protein in normal colorectal mucosa tissue,colorectal adenomas tissueand colorectal cancer tissue suggestes that K-ras is related to the development of colorectal adenomas tissue and colorectal cancer tissue.2.The relationship between HDAC6 and colorectal adenomas was not significant,but HDAC6has a relationship with the occurrence and development of colorectal cancer.The expression level of HDAC6 is related to the degree of malignancy of colorectal cancer.3.There is a positive correlation between K-rasand HDAC6 in colorectal cancer tissue.It is indicated that HDAC6 and K-ras may be involved in the development and progression of colorectal cancer.
