Colorectal cancer is one of the most common human malignancies.Its morbidity and mortality respectively ranks the third and fourth in all malignant tumors.It greatly jeopardizes patient's health and causes a substantial burden to his family.A great part of patients have lost the opportunity for surgery when they first searching for medical consult.Because of the high morbidity and poor prognosis of colorectal cancer,especially for advanced colorectal cancer,further exploring its pathogenesis is very important,and it will provide a new therapeutic direction for the disease.Like other malignant tumors,the occurrence of colorectal cancer is usually caused by oncogene activation and tumor suppressor gene inactivation.But till now,the biological mechanisms of tumor gene regulation are still in research stage.Since Lee and his colleagues for the first time found miRNA,the mechanism of miRNA in vital movement has been excavated,especially in the occurrence and development of tumor.MiRNA is one of the highly conservative and non-coding small RNAs,which controls about a third of the human gene expression.MiRNA usually promotes or inhibits the occurrence and development of tumor by control the signal pathway of oncogene or tumor suppressor gene,thus plays an significant role in the diagnosis,treatment and prognosis of tumor.MiRNA-101 as a member of miRNA family has a reduced expression in many human tumors,inhibits the proliferation of cancer cells,and plays as tumor suppressor gene in the occurrence and development of tumor.Some previous studies have found that miRNA-101 had a low expression in colorectal cancer and it could inhibit the proliferation and metastasis of cancer cells by acting on different target genes.But most of these studies are in vitro studies,and have less clinical samples.So,based on the previous studies,our research increased the clinical sample size,then analyzed the expression of miRNA-101 in colorectal cancer and colorectal adenomas polyp,and reveal its role in the process of colorectal cancer.Our research may help to provide a new approach for the diagnosis and treatment of colorectal cancer.Objective: Our study will investigate whether the expression of miRNA-101 is different in colorectal cancer and colorectal adenomas polyp,then discuss the role of miRNA-101 in the progress of colorectal cancer.Methods : miRNA-101 level was measured in 33 patients with colorectal adenomas polyp and paired non-polyp tissues(including 8patients with tubulovillous adenoma and 25 patients with tubular adenoma)and 31 patients with colorectal cancer and paired non-tumor tissues by real-time PCR.The relative expression of miRNA-101 was analyzed by SPSS 22.0 software.Because the expression of miRNA-101 was skewed distribution,so we used Wilcoxon Matched-Pairs Signed-Ranks Test andTwo samples rank sum test.The difference will be statistically significant when P<0.05.Then we compared whether the expression of miRNA-101 was different in colorectal adenomas polyp and colorectal cancer,analyzed the relationship between the expression and age,gender,pathological type and other clinical significance.Finally,we analyzed the role of miRNA-101 in the occurrence and development of colorectal cancer.Results:(1)the expression of miRNA-101 in colorectal adenomas polyp tissues is higher than in paired non-polyp tissues(P<0.05);(2)the expression of miRNA-101 in patients with colorectal adenomas polyp has no significant difference with respect to ages,gender and pathological types(tubulovillous adenoma and tubular adenoma);(3)the expression of miRNA-101 in colorectal cancer tissues is lower than in paired non-tumor tissues(P<0.05);(4)the expression of miRNA-101 in patients with colorectal cancer has no significant difference with respect to ages,gender and tumor locations.Conclusions:The expression level of miRNA-101 increased in colorectal adenomas polyp and decreased in colorectal cancer,which revealed that miRNA-101 played an important role in the progress of normal colorectal tissue — colorectal adenomas polyp — colorectal cancer.Our study has provide the experimental basis for miRNA-101 to be an indicator of colorectal cancer prediction and prognosis.
