Study On Effect About Chaihu-shaoyao Compatibility Co-nsisted In Prescription Sinisan On TGF-?1 And Other Indicators In Liver Organization Of Immune Hepatic Fibrosis Rats

?Objective?To study rats model with liver fibrosis induced by un-inactivated pig serum,to study the effects of Prescription Sinisan different dose group?high,medium,low and preventative?,Chaihu-Shaoyao different dose group?high,medium,low and preventative?,Colchicine contrast group on TGF-?1,AST and ALT,in order to research Chaihu-Shaoyao compatibility consisted in Prescription Sinisan how to take effects on antifibrotic action,proper dose and the preventive further on the basis of thought and method in the past years.?Methods?One hundred and ten SPF-Winstar rats,120-130g,were randomly divided into eleven groups:control group,model group,colchicines contrast group Sinisan different dose group?high,medium,low and preventative?Chaihu-Shaoyao different dose group?high,medium,low and preventative?.all the other groups were injected with un-inactivated pig serum into abdominal cavity except normal group,0.5ml every rat each time and twice per week,lasting10 weeks.The preventative groups were treated with corresponding drugs when it is began?Sini San preventative group 0.4g/100g and Chaihu-Shaoyao preventative group0.2g/100g?,others groups were treated with their drugs after 6th week.Sinisan different dose group?high,medium,low;1.4g/100g,0.7g/100g,0.35g/100g?Chaihu-Shaoyao different dose group?high,medium,low;0.7g/100g,0.35g/100g,0.17g/100g?colchicines control group?0.2mg/kg?,control group and model group were perfused with equivalence saline.Observing cell morphology transformation and indexes in liver tissues of each group by HE and Masson staining method,immunohistochemical staining method at the end of the experiment.?Results?Prescription Sinisan group could decrease the content of TGF-?₁,AST and ALT.Compared treating groups with model group,there was obvious difference?P<0.05?;The Chaihu-Shaoyao group was better than Model group?P<0.05?,and there was obvious difference between Prescription Sinisan group and Chaihu-Shaoyao group?P<0.05?.Both?high,medium?dose group of Prescription Sinisan and Chaihu-Shaoyao were better than themselves low-dose?P<0.05?Compared high-dose group with medium-dose group,there was no obvious difference?P>0.05?.Preventative-dose group is better than medium-dose group or colchicines contrast group,there were obvious difference?P<0.05?.Sini san medium-dose group was obvious difference compared with Chaihu-Shaoyao group?P<0.05?.The expression of TGF-?₁ in the liver tissues in treatment groups significantly decreased as compared to that in hepatic fibrosis group.?Conclusion?Both Prescription Sini San and Chaihu-Shaoyao compatibility have a positive action of treatment on hepatic fibrosis model rats.Compared to Prescription Sini San,curative effect of Chaihu-Shaoyao compatibility is poor.medium-dose group has a better curative effect while that of the prevention group in Prescription Sini San is outstanding.
