【Objective】To observe and research high-dose group, medium dose group and low-dose groupof supplemental Sini San, Colchicin contrast group, prevention group,Sini San controlgroup how to affect smads,P-STAT3,P38MAPK and a-SMA in hepatic fibrosisrats.Through this study to investigate the mechanism of antifibrotic action and properdose and the preventive action of supplemental Sini San.【Methods】Eighty Wistar rats were divided into eight groups: control group,modelgroup,colchicines control group(0.2mg/kg), prevention group(0.7g/100g),high dosegroup(1.4g/100g),medium dose group(0.7g/100g),and low dose group(0.35g/100g)of supplemental Sini San, Sini San control group(0.4g/100g). Except the control group,all the other groups were injected with pig serum without inactivation into abdominalcavity, twice per week and0.5ml each time,lasting10weeks.The prevention groupwas treated with supplemental Sini San,10weeks at the same time when establishmentof the models began, the other groups were treated with corresponding drugs after6weeks of modeling, to the end of the10week.The changes of the indexes of liverfibrosis in serum were detected respectively.【Results】Supplemental Sini San could decrease the content of SMAD2、SMAD3、P-STAT3、p38MAPK and a-SMA obviously.Compared treating groups with modelgroup,there was obvious difference(P<0.05);The mediate-dose group was better thanlow-dose(P<0.05), and there was obvious difference(P<0.05). The high-dose groupwas better than low-dose(P<0.05), and there was obvious difference(P<0.05), but therewas no obvious difference between high-dose group and mediate-dose group (P>0.05).Supplemental Sini San prevention group was better than mediate-dose group andColchicin contrast group, and there was obvious difference(P<0.05). Moreover, there was obvious difference between Supplemental Sini San mediate-dose group and SiniSan contrast group(P <0.05).The expression of SMAD7in the liver tissues in treatmentgroups significantly increased as compared to that in hepatic fibrosis group.【Conclusion】Supplemental Sini San has a positive action of treatment on hepatic fibrosis modelrats. Mediate-dose group has a better curative effect, the curative effect of theprevention group is best.
