Objective:Low-intensity pulsed ultrasound(LIPUS)Is a longer wavelength of sound waves,The range of frequency is 20 k Hz-1 MHz.LIPUS has a significant effect on repairing the soft and hard tissue injury,with noninvasive,painless,no side effects,easy to penetrate in the organization,good direction and promote drug penetration and absorption advantages.Triamcinolone acetonide has a strong and lasting anti-inflammatory,anti-allergic effect.In this study,LIPUS and triamcinolone acetonide were used in combination with golden hamster buccal mucosal ulcer,record the healing time of oral ulcer and the contents of Superoxide Dismutase(SOD)and Malondialdehyde(MDA).Evaluate the effect of low-frequency ultrasound therapy on oral mucosal ulcer in Syrian Hamster combination with triamcinolone acetonide and the mechanism.Methods: A Sixty Syrian Hamsters were randomly divided into five groups,including a baseline group(contains a normal group and a model group,n=6)and four experiment groups(LIPUS processing and drug using group,LIPUS group,drug group and a normal control group without any processing,n=12).Four experiment groups and the model group were given oxygen free radicals to model the oral mucosal ulcer.Then the baseline group were executed to test SOD and MDA immediately.At 24 h after last treatment,the healing time of ulcer,the contents of SOD and MDA were compared between each group.Residual golden hamsters were recorded healing time,SOD and MDA after the ulcer healed.Results:Compared with LIPUS group,drug group and control group,the healing time of oral mocosal ulcer in LIPUS and drug combined group was shortened.At 24 h after last treatment,the activity of SOD shows that the LIPUS and drug combined group[(2.32±0.30)U/mgprot] were significantly higher than the LIPUS group[(1.83±0.15)U/mgprot],the drug group[(1.76±0.25)U/mgprot] and control group[(1.71±0.18)U/mgprot](P<0.05).The content of MDA shows that the LIPUS and drug combined group[(8.17±0.21)nmol/mgprot] were significantly lower than the LIPUS group[(9.00±0.44)nmol/mgprot],the drug group[(9.04±0.43)nmol/mgprot] and control group[(9.03±0.46)nmol/mgprot](P<0.05).After the oral mucosal ulcer healing,the activity of SOD shows that the LIPUS and drug combined group[(3.33±0.43)U/mgprot] were not significantly different from the LIPUS group[(3.41±0.55)U/mgprot],the drug group[(3.26±0.53)U/mgprot] and control group[(3.40±0.49)U/mgprot](P>0.05).The content of MDA shows that the LIPUS and drug combined group[(7.25±0.47)nmol/mgprot]were not significantly different from the LIPUS group[(7.40±0.35)nmol/mgprot],the drug group[(7.29±0.49)nmol/mgprot] and control group[(7.62±0.30)nmol/mgprot](P>0.05).Conclusion:It is stable and reliable to induce the oral mucosa ulcer by“reactive oxygen species induction”method.In this study,low frequency ultrasound combined with triamcinolone acetonide can significantly shorten the healing time of experimental oral ulcers,effectively increase the activity of SOD,reduce the content of MDA,and accelerate the process of ulcer healing.
