Objective:This study aims to explore the possible pathogenesis of cognitive disorder for SD rats overexpressing MicroRNA-30 e and to observe the effects of Modified-buwang-powder on learning and memory of such model rats.Methods:Forty healthy male SD rats(200g±10g)were randomly equally divided into five groups: blank control group(B),empty lentivirus particle control group(C),overexpressing MicroRNA-30 e model group(M),low-dose Modified-buwang-powder group(L),high-dose Modified-buwang-powder group(H).1ul lentivirus particle carrying the microRNA-30 e was injected into bilateral hippocampus of the rats in group M,group L,and group H respectively,and 1ul empty lentivirus particle was injected into bilateral hippocampus of the rats in group C respectively,while 1ul normal saline was injected into bilateral hippocampus of the rats in group B respectively.After two weeks,the rats in group H and L were given high-dose(0.3g/100g)and low dose(0.15g/100g)Modified-buwang-powder respectively by gavage,the other three groups of rats were given normal saline.Morris maze was used to evaluate the ability of learning and memory in all rats two weeks later.Then the brain tissue of all rats was taken out,and bilateral hippocampal tissue was separated to prepare the tissue homogenates.The activities of ChAT were detected by colorimetry.Results:1.Results of Morris water maze test : Compared with group B,there was no significant difference in the indexes of the escape latent period of the rats,the number of crossing platform and the time of staying at the original platform quadrant with group C(P>0.05).Compared with group B or C,he escape latent period of the rats in group M was significantly prolonged from the second day to the fifth day(p<0.05),the number of crossing platform was significantly decreased(p<0.05)and the time of staying at the original platform quadrant was significantly shorter(p<0.05).Compared with group M,the escape latent periods in group H and L were significantly shorter from the second day to the fifith day(p<0.05),the number of cross platform was significantly increased(p<0.05)and the time staying at the original platform quadrant was significantly prolonged(p<0.05).Compared with group L,the escape latent period of the rats in group H was significantly shorter from the second day to the fifith day(p<0.05),the number of crossing platform was significantly increased(p<0.05)and the time staying at the original platform quadrant was significantly prolonged(p<0.05).2.Results of ChAT activity: Compared with group B,there was no significant difference in the ChAT activity in hippocampus with group C(P>0.05).Compared with group B or C,the ChAT activity in hippocampus was significantly decreased in group M(p<0.05);Compared with group M,the ChAT activities in hippocampus of group H and L were all significantly increased(p<0.05).Compared with group L,the ChAT activity in hippocampus was significantly increased in group H(p<0.05).Conclusion:1.Modified-buwang-powder could improve learning and memory ability of the overexpressing microRNA-30 e SD rats dose-dependently.2.The pathogenesis of cognitive disorder in overexpressing microRNA-30 e SD rats is related with the decrease of ChAT activity in hippocampus,while Modified buwang-powder could improve it dose-dependently.
