Objective:1.To observe the effects of Modified-buwang-powder on learning and memory in AD model rats.2.To observe the effects of Modified-buwang-powder onin vivo hippocampal Long-term potentiation in AD model rats.Methods:40 male Wistarrats wererandomlydivided into 4 groups:control group,AD model group,low-dose Modified-buwang-powder group,high-dose Modified-buwang-powder group. The later three groups were duplicated to AD model by taking intraperitoneal injection of D-galactose for 6 weeks, along with destroying bilateral Meynert nucleus by injection of IBO at the 43 nd day. Low-dose group and high-dose group were given Modified-buwang-powder by gavage from the 1st day for 8 weeks. 8 weeks later, Morris maze was used to evaluate the effects of Modified-buwang-powder on learning and memory, and LTP technology was used to evaluate the effects of Modified-buwang-powder on synaptic plasticity.Results:1. The effects of Modified-buwang-powder on learning and memory:All rats appeared ability of learning and memory after a 5-day training, with escape latency shorter gradually. The mean escape latency of AD model group was significantlylonger than the control group(p<0.01).The mean escape latency of low-dose group and high-dose group was significantly shorter than the control group(p<0.05). The mean escape latency of low-dose group was significantly longer than the high-dose group(p<0.05). The mean times of passing platform from the control group,the AD model group,the low-dose group and the high-dose group were respectively 14.89±1.45、4.75±1.91、7.57±1.51、10.71±0.76,which remained statistically significant(p<0.05).2. The effects of Modified-buwang-powder onin vivo hippocampal long-term potentiation:Given HFS(High frequency stimulation) for 60 mins, the f EPSP(Field excitatory postsynaptic potential)amplification of control group, model group, low-dose group and high-dose group respectively reached 192.18±11.21,110.44±7.90,140.61±7.59,161.16±6.70, which remained statistically significant(p<0.05).Conclusions:Modified-buwang-powder could improve learning and memory ability of AD model rats and increase damaged LTP in AD model rats dose-dependently.
