Objective: To observe the effect of MBWP on learning and memory ability of AD transgenic model mice(APP/PS1 mice).To evaluate the effect of MBWP on the expression of apoptosis-related proteins(Bcl-2,Cleaved caspase-3)in APP/PS1 mice,and explore its mechanisms to improve learning and memory.Methods: 1.Grouping and feeding: 6-month-old APP / PS1 mice were divided into there groups,10 mice each group,randomly.There are defined as APP/PS1 group(APP/PS1 group),MBWP low-dose group(APP/PS1+L-MBWP group,15 mg/kg/d)and MBWP high-dose group(APP/PS1+H-MBWP group,30 mg/kg/d),respectively.Ten non-transgenic mice were used as the control group.2.Behavioral testing: After four months of MBWP intervention,mice were subjected by Morris water maze and shuttle-box test to evaluate their learning and memory abilities.3.Western Blot: Use Western blot technology to detect the expression levels of Bcl-2 and Cleaved caspase-3 in the hippocampus of APP/PS1 mice.Results: 1.Behavioral test results:(1)Morris water maze: In the positioning navigation experiment,the escape latency of the mice in each group was shortened as the experimental days increased.In the space exploration experiment,the stay time in the target quadrant of the APP / PS1 group was significantly shorter than that of the Control group(P<0.01);compared with the APP/PS1 group,the APP/PS1+L-MBWP group and the APP/PS1+H-MBWP the target quadrant stay in MBWP group was prolonged(P<0.05,P<0.01).There was no significant difference between the APP/PS1+L-MBWP group and the APP/PS1+H-MBWP group(P>0.05).(2)Shuttle box experiment: In the training stage of the shuttle box experiment,it was seen that the number of active escapes of each group increased with the length of the training days.During the testing period,compared with the Control group,the number of active escapes in the APP/PS1 group was significantly reduced(P<0.01);compared with the APP/PS1 group,the APP/PS1+L-MBWP group and APP/PS1+H-MBWP group increased(P<0.01,P<0.01);compared with the APP/PS1+L-MBWP group,the APP/PS1+H-MBWP group There was no significant difference in the number of mice's active escapes(P> 0.05).2.Western Blot test results:(1)The expression level of Bcl-2 in hippocampus of APP/PS1 mice were significantly lower than those in the Control group(P<0.05);compared with the APP/PS1 group,APP/PS1+L-MBWP and APP/PS1+H-MBWP was significantly increased(P<0.05,P<0.01).Compared with the APP/PS1+L-MBWP group,APP/PS1+H-MBWP group was significantly increased(P<0.05).(2)The expression level of Cleaved caspase-3 in the hippocampus of APP/PS1 mice was significantly higher than that in the Control group(P<0.01);compared with the APP/PS1 group,were significantly reduced(P<0.05),no significant difference in APP/PS1+L-MBWP group(P>0.05);compared with APP/PS1+L-MBWP group,APP/PS1+H-MBWP group mice was no significant difference(P>0.05).Conclusion: 1.MBWP can improve the learning and memory ability of APP/PS1 transgenic mice.2.The expression of anti-apoptotic protein Bcl-2 in the hippocampus of APP/PS1 transgenic mice was decreased,and the expression level of apoptotic executive molecule Cleaved caspase-3 increased,there are involved in apoptosis.3.MBWP could up-regulate the expression of anti-apoptotic protein Bcl-2 in the hippocampus of APP / PS1 transgenic mice,down-regulate the expression of Cleaved caspase-3 protein and inhibit apoptosis.
