Objective: In this study,we established the H22 model of tumor bearing mice,the mice were divided into four groups,treatment drugs including Huazhuo Jiedu Guben prescription,5-fluorouracil and combined medication,the survival status,tumor size,immune status,pathological changes,tumor markers VEGF,MMP-9expression were observed,evaluate the drug effects,and Study on the effect and mechanism of traditional Chinese medicine in the treatment of Primary liver cancer.Methods: The model of H22 tumor bearing mice was established in 40 healthy mice.After modeling success,40 mice were randomly divided into four groups,including model group(M),Huazhuo Jiedu Guben prescription group(Z),5-fluorouracil group(X),Huazhuo Jiedu Guben prescription combined with 5-fluorouracil group(L).And give related drug treatment.The Chinese medicine group was treated with Huazhuo Jiedu Guben prescription 0.2ml.d-1;The Western medicine group was treated with 5-fluorouracil 20 mg.kg-1.d-1 intraperitoneal injection therapy;Combined treatment group was given Huazhuo Jiedu Guben prescription by gavage in the morning,and the 5-fluorouracil intraperitoneal injection in the afternoon;Model group were given 0.4ml.d-1 normal saline by intragastric administration.Treatment for 12 days,observe the coat color,eating condition,mental state,weight change and death situation,calculated with the inhibition rate,spleen index and thymus index,observed the expression of MMP-9 and VEGF by immunohistochemistry.Results:1 The general condition of miceAfter treatment,all mice were not as good as before treatment,especially in the model group,the mice were dispirited,and the coat color was rough,loss of appetite,have no significant increase in body weight,and the tumor was significantly enlarged.Compared with the Western medicine group,the traditional Chinese medicine group and the combined medication group were better than the Western medicine group in the mental state,appetite and weight gain.2 The tumor growth of miceIn the aspect of tumor inhibition,the inhibition rate of the combined group was the highest,and that of the Western medicine group was higher than traditional Chinese medicine group.The tumor weight of the three treatment groups was less than the model group,the difference was statistically significant(P<0.05);The tumor weight of combined group was less than Western medicine group and Chinese medicine group,significant difference(P<0.05);The tumor weight of Western medicine group was less than the traditional Chinese medicine group,the difference was statistically significant(P<0.05).3 The spleen index of miceThe spleen index of Chinese medicine group was the highest in all groups,obviously higher than the other groups,the difference was statistically significant(P<0.05);The spleen index of combined treatment group significantly higher than the Western medicine group and model group,significant difference(P<0.05);Compared with the model group,spleen index of Western medicine group was statistically significant,the difference was not statistically significant(P>0.05).4 The thymus index of miceCompared with the model group,the thymus index of all treatment groups were significantly increased,the difference was statistically significant(P<0.05);The thymus index of the traditional Chinese medicine group and the combined group were significantly higher than those of the Western medicine group(P<0.05);The thymus index of combined group compared with traditional Chinese medicine group,the difference had no statistical significance(P>0.05).5 Expression of VEGF in tumor tissues of miceThe content of VEGF in tumor tissues from all treatment groups were significantly lower than that of model group,the difference was statistically significant(P<0.05);VEGF in tumor tissue of combined group was significantly lower than that of traditional Chinese medicine and Western medicine group,the difference was statistically significant(P<0.05);VEGF in tumor tissue from the Western medicine group was lower than Chinese medicine group,the difference was statistically significant(P<0.05).6 Expression of MMP-9 in tumor tissues of miceThe expression of MMP-9 protein in all treatment groups was significant difference from the model group(P<0.05);The expression of MMP-9 in combined treatment group was lower than Chinese medicine group and Western medicine group,the difference was significant(P<0.05);MMP-9of Western medicine group was lower than that of the traditional Chinese medicine group,the difference was statistically significant(P<0.05).7 Pathological changes of mice tumorThe growth of tumor tissue in model group was vigorous,tumor cell specificity,the color was very deep,alkaline strongly,rich blood vessels microscopically;The growth of tumor tissue in Chinese medicine group was slow,alarge number of tumor cells degeneration,large area necrosis,cytoplasmic and nuclear eosinophilia;The growth of tumor tissue was not strong in the western medicine group,patchy necrosis and vacuoles were found,cell atypia decreased,and the nucleus showed weak acidity;The tumor growth was not obvious in the combination group,partial tumor cell necrosis,cytoplasm and cell nucleus were very acidic,the blood vessels were significantly reduced compared with the model group.Conclusions:1 It is suggested that Huazhuo Jiedu Guben prescription can improve the anti-tumor effect of 5-fluorouracil,inhibit the growth of tumor,and increase the tumor inhibition rate2 Huazhuo Jiedu Guben prescription can obviously improve the immunosuppressive effect of 5-fluorouracil on H22 liver cancer mice,enhance the immune function of HCC mice,and improve the quality of life of liver cancer mice.
