| Objective:After establishing the H22 hepatic cancer mice model by cell vaccination, the cancer-bearing mice were treated by Huazhuo Jiedu Ruanjian Granules or 5Fu as control group. It is that the indexes were observed,which inluding the observation conditions, the general situation,the tumor growth,the related immune index, the VEGF content of serum and the VEGF and MVD’expression in the tumor tissue.To explore the effect in anticancer growth and mechanism of Huazhuo Jiedu Ruanjian Granules in H22 hepatic cancer mice.Method:Inoculate mice abdominal cavity with H22 tumor cells, six days later, aseptic extract ascites after it is generated,it was diluted into cell suspension designated. 50 KunMing mice were randomly divided into 5 groups: model group(M), Huazhuo Jiedu Ruanjian Granules group(Z), Huazhuo Jiedu Ruanjian Granules in combination with 5Fu group(Z+X), 5Fu(X), and Normal control group(N), 10 mice in each group. each mouse’s left axillary the first four groups was inoculated 4×106 cells suspension of H22 tumor, and given the corresponding drug intervention 7 days in a row after 24 hours. Remove eyes of mice before weighing each mice’ weight, and collect blood for detect the serum of VEGF levels by ElASA; After killing mice by broking neck,open abdominal cavity and blunt dissection tumor,spleen, thymus,which were weighed respectively;calculate the inhibitory rate, spleen index and thymus index and detect the expression level of VEGF and MVD of tumor tissues which fixed in 4% neutral formaldehyde with immunohistochemical method.Results:1 The comparison of general condition of the miceCompared wih the normal group mice,tumor-burdened group mice were significantly increased diet intake and activity,they were in depression without burnish fur but with goiter in left axillary.the model group performance was more outstanding. Between the treatment group, Z+X group mice performance was better than the other, X group’ performance was wore than Z group’.2 The comparison of tumor tissue morphological appearance in tumor–burdened groupM group’tumor: shape is irregular, color is grey and red, texture is soft, no obvious capsule, tumor invasion thoracic, and even lung tissue; Z group’tumor: dead colour is not uniform, some were dark yellow, toughening, one sample had capsule completely; Z+X group tumor: shape is Small, color was flesh pink, soft. Four samples had capsule completely;X group tumor: dead colour,some were bright red partly, toughening, two samples had capsule completely.3 The comparison of tumor growth in in tumor –burdened group.Between three treatment, Z+X group’ inhibitory rate was highest, the second was the X group, the third was Z group. The average tumor weight of tumor weight Z+X group was minimum, which was followed by the X group, Z group, M group successively.as for the average tumor weight,the three treatment group’ were less than M group’, the difference was statistically significant(P<0.05); Z+X group tumor weight was less than X group significantly(P <0.05); X and Z group, have no difference(P >0.05).4 The comparison of the spleen inde in different groupsCompared with the normal control group, a tumor-burdened group mice’ spleen index increased,the difference was statistically significant(P<0.05); Treatment group were lower than M group significantly(P<0.05); spleen index of Z group and Z+X group was lower than that of X group, the difference was statistically significant(P<0.05); Z group and Z+X group were no significant difference(P>0.05).5 The comparison of thymus index in different groupsCompared with the normal control group,the thymus weight of tumor-burdened groups mice decreased, the difference was statistically significant(P<0.05); Between tumor-burdened groups, the thymus index of M group, Z group, Z+X group were higher than that of X group, the difference had statistical significance(P<0.05); Z group and Z+X group were no significant difference(P >0.05)6 The comparison of the serum of VEGF in different groupsCompared with the normal control group’s, the serum of VEGF in tumor-burdened groups was higher significantly(P<0.05); Between tumor-burdened groups, the treatment group mice were lower than M group, the difference was statistically significant(P<0.05); Between the treatment group, Z group and X group were higher than Z+X group, the difference was statistically significant(P<0.05); Compared with the X group, Z+X group decreased significantly, significantly(P<0.05); X and Z group had no significant difference(P>0.05).7 Comparison of the VEGF expression of tumor-tissue in different tumor-burdened groupsCompared with model group, the VEGF integral optical density(IOD) and positive area of treatment groups were reduced obviously(P<0.05); Comparison among treatment groups, IOD and positive area of Z+X group was less than Z group and X group, there were significant differences(P<0.05); IOD of X group was less than Z group(P<0.05),while positive area, they have no difference significantly(P>0.05).8 Comparison of the MVD expression of tumor-tissue in different tumor-burdened groupsAmong the tumor-burdened groups, the MVD expression of tumor-tissue was difference significantly each group(P<0.05); the Z+X group were lowest, Compared with M group, the treatment groups decreased significantly,(P<0.05); Z+X group is lower than the Z group and X group, the difference was statistically significant(P<0.05); X is lower than the Z group, there were significantly differences(P<0.05).Conclusions:1 Huazhuo Jiedu Ruanjian Granules party can improve general condition in H22 tumor-burdened mice, reduce tumor weight, increase the inhibitory rate.It has shown a protective effect in inhibiting tumor growing.2 Huazhuo Jiedu Ruanjian Granules can improve splenomegaly, reduce the excessive consumption of thymus, adjust immune function in mice.3 Huazhuo Jiedu Ruanjian Granules can reduce the serum of VEGF levels, reduce the MVD expression in tumor tissue, inhibit tumor angiogenesis, thus it Plays a role in inhibiting the tumor growth and metastasis in mice. |
