Objective: To investigate the influence and mechanism of Psammosilene tunicoides on the expressions of IL-1?,MMP-3 and TIMP-1 in plasma and inflammatory factors of articular cartilage with knee osteoarthritis(OA).Methods: 120 healthy rabbits were randomly and equally divided into blank control group,model control group,Psammosilene tunicoides group,Glucosamine group,each group of 30.Each group was divided into three subgroups according to the time of 4,6,8weeks,each subgroup was a total of 10 rabbits.Except for the blank control group,the others were induced by Hulth's method to copy model of knee OA.The rabbits in the Psammosilene tunicoides group and Glucosamine group were given 12.8 mg/kg/d.The rabbits in the normal group and model group were given with the same amount of normal saline gastric(10 m L)one time per day,over a 8-week period.At 4,6,8 weeks after operation were randomly selected in each group 10 rabbits underwent venous blood of rabbits and killed by gas embolism.Right knee femoral condyle and tibial plateau for observation and measurement.Mankin score was used to evaluate the degeneration of articular cartilage.The m RNA relative expression level of IL-1 ?,MMP-3,TIMP-1 in articular cartilage were detected by RT-PCR.ELISA method was used to detect the protein expression levels of IL-1?,MMP-3 and TIMP-1 in rabbit plasma,and to calculate the MMP-3/TIMP-1 value.Results: With the extension of 4,6,8 weeks,the Mankin score of articular cartilage in each group increased gradually,which indicated that the progress of OA increased with time.The Mankin score of the model group was higher than that of other groups,and the difference was statistically significant(P<0.05).The results showed that the improved Hulth method was successful in replicating OA animal model.The expression of IL-1?,MMP-3,TIMP-1 m RNA and protein in the model group at each time point of OA was higher than that in the blank control group,the difference was statistically significant(P<0.05).In reducing IL-1?m RNA and protein expression,the role of the Psammosilene tunicoides was equivalent to Glucosamine,the difference was no statistically significant(P> 0.05);When reducing the expression of MMP-3 and increasing the expression of TIMP-1 m RNA and protein,the effect of Psammosilene tunicoides was significantly higher than that of Glucosamine,the difference was statistically significant(P <0.05).Psammosilene tunicoides in regulating MMP-3 and TIMP-1 balance than Glucosamine obviously,the difference was statistically significant(P< 0.05).Conclusion: Psammosilene tunicoides can reduce the levels of expression of IL-1 ?and MMP-3 and can increase in the expression level of TIMP-1 in knee OA,the factor of degeneration of articular cartilage was controlled to protect articular cartilage,thus delaying the progression of knee OA.
