Objective: This article mainly aims to explore the curative effect by using Joint I herb soup for curing Rat’s Knee Osteoarthritis and the influence on expression of MMP-3/MMP-13 and TIMP-1 in Articular Cartilage Tissue;Method: 9 Wistar rats were randomly selected as normal group, and the remaining 45 were chosen to establish model, referring to Hulth model, after get the obvious KOA model, the rats are randomly divided into 5 group(8 rats each one),and named each group as model, low dose of Joint 1, middle, high and positive control group. According to experimental design,Rats are treated by drug intervention. During this period, we should observe and record the status of the joints and the rats’ behavior changes before and after the test for each group.After 6 weeks, kill the rats and test the index:(1) The eyes observation and score of cartilage surface: After the rats were killed, their right knee joints were cut, and three trained workers were chosen to observe the general performance of articular cartilage by naked eyes according to the Principle of Double Blind, and scored according to the standard of literature;(2) Make the general optical microscope slides by using the extracted Cartilage tissue, observe the pathological changes and then score according to Mankin’s Articular Cartilage Histological Scoring System;(3) Make the section by extracting the Articular cartilage tissue, and test the expression of MMP-3/MMP-13 and TIMP-1 according to Immunohistochemistry;(4) Test the expression of MMP-3/MMP-13 and TIMP-1 by using the RT-PCR Technology.Results:(1) Before establishing model, the activity and form of all rats’ right rear knee joints are normal,after establishing model, all groups, except for the normal one, have some KOA variations, such as inflating of right posterior knee joint, inversional deformity, limping and poor joint activities. And after medication, there is no change for the normal group’s Right posterior knee joint, but the others have different KOA changes, which is less than the modeling groups.(2) The results from eyes scores show that the model and treatment groups are higher than the normal one; the scores of low, middle and high dose of Joint 1, and positive control group are lower than the model group, and the middle dose of Joint 1 and positive control group are lower than the low one. All above difference has statistical significance(P<0.05).(3) The results from Mankin’s score show that the model and treatment group is higher than the normal group, the low, middle and high dose of Joint 1, and positive control group are lower than the model group, and the middle dose of Joint I and positive control group are lower than the low one. All above difference has statistical significance(P<0.05).(4) The results from Immunohistochemistry show that the right posterior knee joints of the normal group have a small MMP-3/MMP-13 and TIMP-1 positive expression; the expression of MMP-3/MMP-13 and TIMP-1 for the model and treatment group is higher than the normal group; the expression of MMP-3/MMP-13 of the low, middle and high dose of Joint 1, and positive control group are lower than the model group,TIMP-1 is higher than it; the expression of MMP-3/MMP-13 of the middle dose of Joint 1 and positive control group are lower than the low group, and TIMP-1 is higher than them. All above difference has statistical significance(P<0.05).(5) The results from q RT-PCR test show that the expression of MMP-3/MMP-13 gene expression quantity for model group is higher than the normal group; the expression of MMP-3/MMP-13 gene expression quantity for the low, middle and high dose of Joint 1, and positive control group is lower than the model group, and TIMP-1gene expression quantity is higher than it; the expression of MMP-3/MMP-13 gene expression quantity for the middle and high dose of Joint 1 and positive control group are lower than the low one, and TIMP-1gene expression quantity is higher than it;...
