Investigating The Neuroprotective Effect Of Tianmagouteng Drink For Rats With Parkinson's Disease Induced By 6-OHDA Based On Autophagy

Objective: Application of tianmagouteng drink on 6-6-hydroxydopamine(6-OHDA)induced by Parkinson disease(Parkinson s disease,PD)rats,to study the effect of tianmagouteng drink on the behavior of PD model rats;observation of dopamine in brain tissue of rats in the PD model(DA)were detected,the protective effect of tianmagouteng drink on DA neurons in PD rats;to observe the expression of LC3 B and Beclin1 in striatum of rat protein,clear tianmagouteng drink on autophagy possible regulation mechanismMethods: 61 male SD 9 rats as normal group,more than 52 models,the model of success(36)were randomly divided into 4 groups: model group,tianmagouteng drink high dose group,low dose tianmagouteng drink group,Madopar group,a total of 5 groups.Intragastric gavage for 8 weeks,before and after the behavioral test,impact test tianmagouteng drink on the behavior of PD model rats;DA rats were sacrificed and the content of brain tissue was detected by ELISA,to observe the protective effect of tianmagouteng drink on DA neurons in PD rats;brain tissue,expression by Western blot method to detect the LC3 B and Beclin1,from the point of view to investigate the effects and mechanism of autophagy,tianmagouteng drink of PD model ratsResult:(1)Effects of tianmagouteng drink on 6-OHDA induced behavioral changes in rats with PDRotation test: after intraperitoneal injection of APO,10 minutes to 40 minutes,there were different degrees of rotation,the number of turns was more than 7 laps,/min rats were 36.Swimming test: Rats swimming time shortened in the model group,compared with the normal group,the difference was statistically significant(P<0.05);compared with the model group,tianmagouteng drink high dose group,low dose tianmagouteng drink group and Madopar group rat swimming time increased,the difference was statistically significant(P<0.05);the Madopar group is higher than that of tianmagouteng drink dose group,low dose group has significant effect,high dose of tianmagouteng drink group is more obvious than tianmagouteng drink low dose group(P<0.05).Suspension experiment: model group rats were suspended for a short time,compared with the normal group,the difference was statistically significant(P<0.05);compared with the model group,tianmagouteng drink high dose group,low dose tianmagouteng drink group and Madopar group rats suspension time prolonged,the difference was statistically significant(P<0.05);the tianmagouteng drink high dose group with Madopar the effect of low dose group,tianmagouteng drink group,Madopar group is better than the low dose of tianmagouteng drink(P<0.05).(2)the effect of tianmagouteng drink on DA content in striatum of PD ratsCompared with the normal group,decrease the content of DA PD group rats,the difference was statistically significant(P<0.05);compared with the model group,tianmagouteng drink high dose group,low dose tianmagouteng drink group and Madopar group rats DA content has increased to varying degrees,the difference was statistically significant(P<0.05);among them,the high dose of tianmagouteng drink compared with Madopar group and tianmagouteng drink low dose group increased significantly,indicating that tianmagouteng drink can increase the content of DA.(3)The effect of tianmagouteng drink on LC3 B and Beclin1 in PD ratsCompared with the normal group,model group,LC3 B expression decreased,Beclin1 expression decreased,the difference was statistically significant(P<0.05);compared with the model group,tianmagouteng drink high dose and low dose tianmagouteng drink group and Madopar group,LC3 B and Beclin1 expression were increased slightly,the difference was statistically significant(P<0.05);the tianmagouteng drink high dose group compared with Madopar tianmagouteng drink group,low dose group has significant effect,high dose of tianmagouteng drink group is more obvious than tianmagouteng drink low dose group(P<0.05)Conclusion:First of all,we used 6-OHDA rat model to observe the behavioral changes of PD rats before and after administration.The results showed that tianmagouteng drink had a certain alleviation effect on the symptoms of PD model rats with slow motion and tremor.In addition,we detected the DA content in the PD rat model,and the results showed that tianmagouteng drink could increase the DA content.Finally,we also examined the expression of LC3 B and Beclin1 in the brain tissue of PD rats,and speculated that tianmagouteng drink could activate autophagy and alleviate PD symptoms in rats by elevating the expression of LC3 B and Beclin1.
