Objective:To obtain the gene diversity and haplotype diversity of the 27 Y-STR loci in Han population from Zhenxiong,Hui population of Weishan,Yunnan Province and Uygur population from Xinjiang,which could enrich the basic data of population genetics,provide data support for forensic identification,promote the establishment of regional Y-STR database and provide data reference for human genetics research.Methods:Blood samples of unrelated male individuals were collected from Yunnan Provincial Public Security Department,including 254 samples from Han population Zhenxiong,171 samples from Hui population,Weishan and 194 from samples from Uygur population,Xinjiang.DNA was direct amplified by using Yfiler(?)Plus system and genotyped by ABI3500 Genetic Analyzer.Data statistical analysis was applied by DISPAN and MEGA4.0,the gene frequency and haplotype frequency distribution of the three populations was analyzed;the different gene and haplotype polymorphism was compared;the DA distance and genetic relationship between three populations was analyzed.Results:We have got the gene frequency and haplotype frequency distribution of the three populations.In Han population from Zhenxiong,the gene frequency of 27 Y-STR loci was from 0.0039 to 0.7244,the gene diversity was from 0.4131 to 0.9548,252 different haplotypes were observed once,1 haplotype occurred twice in 254 samples,the haplotype diversity was 0.999968877;in Hui population from Weishan,the gene frequency of 27 Y-STR loci was from 0.0058 to 0.7207,the gene diversity was from 0.4314~0.9408,167 different haplotypes were observed once,2 haplotype occurred twice in 171 samples,the haplotype diversity was 0.999862401;in Uygur population from Xinjiang,the gene frequency of 27 Y-STR loci was from 0.0052 to 0.6391,the gene diversity was from 0.4131~0.9548,192 different haplotypes were observed once,1 haplotype occurred twice in 194 samples,the haplotype diversity was 0.999946584,there was no shared haplotype among three population.Genetic distance and phylogenetic tree showed that the genetic distance between Han population from Zhenxiong and Taiwanese population was the nearest,the genetic distance between Han population from Zhenxiong and Yi population was the farthest;The Weishan Hui population and Xinjiang Uygur population clustered into a cluster,which the genetic distance between Wa population from Yunnan was the farthest.Conclusion:From the analysis of the three poplations,we have obtained the genetic basis data of 27 Y-STR genes of three poplations,which could be used to provide data support for the establishment of Y-STR database;the 27 Y-STR loci have high genetic polymorphism and haplotype polymorphism,which could be applied(?)on the Han}poplation from Zhenxiong,Hui population from Weishan,Uygur population from Xinjiang,and could be used for the reasearch of forensic science practice and genetics research in three poplations.The RM-STR(DYS449、DYS518、DYS570、DYS576、DYS627、DYF387S1a/b)in the Han poplation from Zhenxiong,Hui population fromWeishan,Uygur population from Xinjiang both have high genetic poly(?)orprphism,theresults suggested that when filtering for Y-STR loci in different regions and different ethnic groups in China,the rapid mutation Y-STR locus with high genetic polymorphism could be used appropriately,from which the genetic data obtained can more accurately reflect the genetic information characteristics of the population.From genetic distance and phylogenetic tree analysis,there were different genetic characteristics between three different regions and different nationalities,the genetic distance between Han nationality in Zhenxiong,Taiwan and Han nationality in Zhejiang was close,showing the genetic characteristics of the Han population in the South;the genetic distance between Hui people in Weishan and Xinjiang Uygur people was the closest,which illustrating that the two ethnic groups have some similarity in paternal inheritance.
