Objective: The epidemiological investigation was performed in the Hetian Area toreport the epidemiological characteristics of lipids levels in Uygur Xinjiang population. Toinvestigate the relationship between the genetic polymorphisms of the Mitogen-activatedprotein kinase activated protein kinase2gene and lipids levels in Uygur Xinjiangpopulation. Methods:350Uygur males and595females were recruited randomly fromHetian area. The2single nucleotide polymorphisms (rs44890, rs45514798) in MK2genewere selected and genotyped by Taqman-PCR in the recruited945Uygur subjects. Allsubjects underwent questionnaire, physical examination, measurement of lipid levels andplasma TNF-alpha.The sequences of MK2gene functional region were sequenced inUygur Xinjiang population with dyslipidemia.7single nucleotide polymorphisms (SNP)in the MK2gene were identified. Results: In the two representative variations genotyped,rs44890and rs45514798, the dyslipidemia and control groups were no significantdifferences of genotype distribution between dyslipidemia and control subjects in twopolymorphisms(P>0.05). In ANCOVA analysis,we found: In males, the concentration oftotal cholesterol(TC)[TT:CT:CC:(4.35±1.20) vs.(4.69±1.34) vs.(4.83±1.44),P=0.033] andTNF-alpha [TT:CT:CC:(106.63±62.39) vs.(128.44±86.15)vs.(153.06±82.99),P=0.001]were different in3genotypes of rs44890,dominant model show that in males theconcentration of TC[TT+CT:CC:(4.72±1.36)mmol/L vs.(4.35±1.20)mmol/L,P=0.012] wasdifferent in CC/CT+TT genotypes of rs44890,in females the concentration oftriglyceride(TG)[TT+CT:CC:(1.45±0.84)mmol/Lvs.(1.64±1.15)mmol/L,P=0.021]wasdifferent in CC/CT+TT genotypes of rs44890.Adjusting for age,BMI,drinking andsmoking in a logistic regression analysis,we found in males TT genotype of rs44890maybe risk factor of high TC[TT vs.CC:OR=2.112(95%CI1.021-4.37,P=0.040)],but need further study. Conclusion: There are no association of the two polymorphisms (rs44890and rs45514798) of MK2gene with lipids levels in Xinjiang Uyghur population.
