Objective To investigate the association between suppressor of cytokine signaling3(SOCS3) geneticpolymorphisms and insulin resistance (IR) in Xinjiang Uygur population.Combining with the phenotypicdata, the possible suspectable marker of IR in Uygur was determined.Methods In this cross-sectional study on the metabolic diseases (e.g. obesity) among Uygur Chinese inHetian, Xinjiang China,1292Uygur individuals were enrolled.The sample size for IR subjects[homeostasismodel assessment for insulin resistance(HOMA-IR)》2.96] was323,whereas that for non-IR controls was969(HOMA-IR<2.96). Genomic DNA was extracted from the peripheral blood by using QIAGEN DNAKit. Representative variations were selected according to gene database and genotyped using the TaqManpolymerase chain reaction method in1292Uygur individuals.A relatively isolated general population in arelatively homogeneous environment and a case-control study was conducted to test the associationbetween the genetic variations of SOCS3gene and insulin resistance.Results (1) There was no significant difference of genotype distribution of rs4969168, rs9914220andrs12953258between insulin resistance and control groups in the total and female population(P>0.05).There was significant difference of genotype distribution of rs4969168between insulin resistance andcontrol groups in the male population(χ2=7.216,P=0.027).(2) Although the insulin resistance relatedquantitative phenotypes have no significantly difference in individuals with GG、AG and AA genotypes ofrs4969168in total、male and female population(P>0.05),the mean of body mass index and the median offasting insulin increased in individuals with GG、AG、AA genotypes of rs4969168in male population.Butnot in total and female population.(3) There are no significant difference of haplotype between insulinresistance and control groups in the total and female population (P>0.05), but haplotype2(rs12953258C-rs4969168A-rs9914220C) was significantly associated with a higher prevalence of IR inmale population (P=0.023).(4) The logistic regression analysis adjusted confounders(such as age,gender)showed that AG genotype of rs4969168variation might be a risk factor for insulin resistance inmale(OR=1.772,95%CI:1.081-2.906,P=0.023).Conclusions The present study suggests that the rs4969168polymorphism in SOCS3gene may beassociated with insulin resistance in Xinjiang Uygur men.
