Objectives:?To observe the expression of visfatin and omentin in different positions and different levels of placenta in normal pregnancy and hypertensive disorder complication(HDCP),and the placental heterogeneity's effect on the expression of visfatin and omentin.?To investigate the role of visfatin and omentin in the pathogenesis of HDCP and their predictive value for HDCP.Methods:53 patients with HDCP were diagnosed at the First Affiliated Hospital of Kunming Medical University,including 26 patients with gestational hypertension(GH),7 patients with mild preeclampsia(PE),and 7 with severe preeclampsia.And 53 cases of normal full-term pregnant women as the control group.All subjects were drawn within 30 minutes after delivery of the placenta and avoided the hemorrhage,infarct and calcification area.The placenta was divided into three groups:center,middle and border.Then the three groups were divided into three layers:the mother surface layer,the villi layer and the fetal surface layer,each size is about 1cm×1cm×1cm.Putting them in the sterilized tube into the refrigerator at-80?.The frozen samples were thawed before the test,and the samples were kept at a temperature of 2-8? after melting.The remaining specimens were transferred to the-80?refrigerator for further storage.The samples were cut and weighed,and 1 g of the sample was added to 500 u1PBS(pH = 7.4)for diluted.The cells were homogenized with a micro-electric homogenizer and centrifuged for 20 minutes(3000 rpm)at a low temperature and high speed centrifuge.Then we carefully collecting cell supernatant.The levels of visfatin and omentin were detected by enzyme-linked immunosorbent assay(ELISA).The optical density value was measured by the microplate reader and the results were analyzed by software.The data were analyzed by SPSS21.0 and were expressed as mean ± standard deviation(X±s).The t-test was used between the two groups and one-way ANOVA was used to compare the four groups.The correlation between the two variables was analyzed by Pearson correlation analysis.Results:? The visfatin and omentin were expressed in placental tissue of hypertensive disorders complicating pregnancy group and control group,and both were expressed in different positions and different levels of placenta.The concentration of visfatin in the control group was 116 ± 99 pg/ml,175 ± 137 pg/ml in the hypertensive group,176±106 pg/ml in the mild preeclampsia group and 179 ±109 pg/ml in the severe preeclampsia group.The content of visfatin in placental tissue of HDCP group increased,and the results of comparison between the four groups showed that the level of visfatin were incresed in the placenta tissue of the GH group,mild PE group and severe PE group comparing to control group(P<0.01),but there was no significant difference between the three groups(P>0.05).? The concentration of omentin in the placenta of the control group was 393 ± 223 ng/L,389± 272 ng/L in the hypertensive group,323 ± 180 ng/L in the mild preeclampsia group and 242 ? 202 ng/L in the severe preeclampsia group,and the levels of omentin were decreased in placental tissue of HDCP patients comparing to control group.The omentin content in placental tissue of GH group was lower than that of control group,but the difference was not statistically significant(P>0.05).The content of placental omentin in mild PE group was significantly lower than that in GH group and control group.The level of placental retina in severe PE group was significantly lower than that in mild PE group,GH group and control group(P<0.05).?In the control group,the content of visfatin in different positions and different levels of placental tissue were 100 ± 80,114 ± 97,119 ± 95,123 ± 103,120 ± 102,131 ± 97,117 ±84,117±97,105± 93 pg/ml,the content of omentin were 433±322,402 ± 298,412 ± 300,382± 226,374 ±217,442±238,426 ± 238,381±198,385±218 ng/L;In the test group,the content of visfatin in different positions and different levels of placental tissue were 182±156,164 ±145,195±156,166 ±147,187±132,207±164,159 ±148,172±154,182±167 pg/ml,the content of omentin were 364±276,324±160,340± 243,317 ± 256,331 ± 239,348± 243,294 ± 213,282 ? 212,324 ± 180 ng/L.In the experimental group,the content of visfatin in different positions and different levels of placenta was higher than that of the control group,while the omentin content was lower than that of the control group.There were no significant differences in the expression of visfatin and omentin between the different tissues of the same patient(P>0.05),but the expression of visfatin and retina in the control group,GH group and PE group were different.?There was no correlation between the expression of visfatin and omentin in placental tissue of HDCP patients.Conclusions:? Compared with normal pregnant women,the expression of visfatin in placental tissue of patients with hypertensive disorder complicating pregnancy was significantly higher and the expression of omentin was lower than that of normal pregnant women.?There were significant differences in the expression of visfatin and omentin between the different positions of placenta and the different levels of placenta in the normal pregnant women,gestational hypertension(GH)and preeclampsia(PE),suggesting that placental heterogeneity can affects the expression of visfatin and omentin.?Visfatin and omentin were important factors in placental tissue of maternal-fetal interface during pregnancy,which may be involved in the development of HDCP,but there was no correlation between them in placental tissue.
