Objective:Consist of herba ephedrae,semen armeniacae amarum and radix glycyrrhizae,the formulae of San'ao Decoction(SAD)first comes from the Taiping Imperial Pharmacy Beneficial Formulas.SAD mainly treats cough that results from exogenous wind-cold syndrome marked by cold by wind pathogenic factor,stuffy nose,heavy voice,cough with profuse,headache and dizziness.Modern clinical medicine uses SAD for the treatment of exogenous wind-cold syndrome caused by acute bronchitis or acute exacerbation of chronic bronchitis.Focusing on SAD as its object of study,this research integrates SAD's ancient literature with clinical and experimental data information;moreover,by enlarging the source of its literature,this research applies data mining methods to the exploration of SAD's clinical application in order to reveal SAD's multi-component,multi-target,multi-channel interactive mechanism.Besides,this research verifies the reliability of data mining results through experiments in the hope of offering objective evidence for SAD's curative effect on asthma and other diseases,and supplementing the existing TCM formulae literature studies with a new approach.Method:1.To review relevant documents about SAD in ancient TCM;to work out SAD's naming process,attending syndromes,prescription characteristics,and modification through traditional literature study methods;to analyze the modern clinical spectrum of disease related with SAD.2.To sort out modern clinical documents about SAD;to apply data mining techniques to the analysis of the clinical application of SAD to asthma from the perspectives of prescription medication,illness,and therapy;to effectively study the information about the drugs,targets,genes of SAD through systems biology methods;to employ Cytoscape V3.1.0 software in network modelling so as to examine SAD's complex multi-component,multi-target,multi-channel network relationship and reveal the possible functioning mechanism and effect approaches of treating asthma with SAD.3.To verify the data mining results through experimentation:MTT method was used to detect the protective effect of the drugs on human bronchial epithelial cells;Hoechst33342 dyeing method was used to observe apoptosis;flow cytometry was used to detect cell cycle and apoptosis,and the percentage of Treg and Th17 cells.Results:1.Results from the systematic review of both ancient and modern literature on SAD:in ancient literature the treatment of the cardinal symptoms involving SAD include nasal congestion,heavy voice,cough,excessive panting,voice loss,and chest distress and shortness of breath;modern spectrum of disease involves over 40 types of clinical diseases,among which those of higher proportions are exogenous cough and bronchial asthma.2.Results from association rules based on frequent item sets mode method:in modern clinical practice SAD is often used with expectorant drugs.Among these expectorant drugs,those of strong support include pinellia,citrus,earthworm,poria,bellflower,perilla,and coltsfoot;the drug pairs of strong support and close compatibility include pinellia-citrus,pinellia-poria,earthworm-perilla,silkworm-cicada,and peucedanum-bellflower;the main effect of SAD is releasing lung,and SAD is combined with other therapy in treating asthma,among which those of relatively greater support include releasing lung,dissipate phlegm,relieve wheezing,invigorating spleen,worming and ventilating lung,dispelling wind and spasmolysis.3.This research collects 5 targets after excluding duplicate data from 22 corresponding targets to the major chemicals of SAD formula via STITCH database.The targets are sorted out in Reactome Human Biology Reaction and Signal Channel Database,and 484 signal channels,including mainly the inflammatory and immune channels,are obtained after the removal of duplicate data.4.Results of the experiments prove that none of the various consistencies of SAD's effective parts are toxic to normal human bronchus epicytes;all of them can protect the cell damage caused by the inflammatory cytokine IL-4;they can also increase the percentage of Treg cells and decrease that of Th17 cells in vitro.The results produced by data mining are proved to be reliable through experiments,and the data mining methods employed in this research are proved to be feasible.Conclusion:This research systematically reviews the descriptions about SAD's attending syndromes and treating principles in both ancient and modern literatures.It studies SAD's modern spectrum of disease.Through the use of frequent item sets mode,this research explores the application characteristics of treating asthma with SAD,and SAD's compatibility characteristics with other drugs via constructing the network of SAD's multi-component,multi-target,multi-channel interactive mechanism.It also verifies the reliability of the results produced by data mining methods.The results of this research prove that to employ data mining techniques in formulae studies not only resolves the problem of massive data and complex drug relationship but enlarges the scale and enriches the research methods of literature study.
