Purposes:To investigate the diseases related to spleen deficiency either in modern medicine or traditional Chinese medicine, as well as the relationship with strength among them based on data mining; Using the method of Association Rules to explore the regularity of medicine compatibility, key medicinal herbs and the characteristics of tonifying spleen prescriptions which from Chinese Prescription Database and some ancient professional books related to the spleen. To evaluate the practicability of above regularity for reference to the clinical therapy of diseases related to spleen deficiency.Material and methods:1. The research articles related to “spleen deficiency” published from 1978 to 2014 were retrieved from Chinese Biology Medicine disc(CBM) and screened to be the analyzing source data.The medical subject headings were extracted from the articles with Bibliographic Item Co-Occurrence Mining System(BICOMB), and the subject headings of disease name were picked up according to the Medical Subject Headings(Me SH) and Subject Headings on Traditional Chinese Medicine. The Tables of diseases highly related to spleen deficiency were generated with word frequency counting.The co-occurrence matrix was generated with BICOMB for the high frenquency disease subject headings. A visualized social network analyzing software, Net Draw, was applied to the analysis of the relationship with strength between spleen deficiency syndrome and the above highly related diseases, and visualized social network maps were generated.2. The tonifying spleen prescriptions from China Prescription Database and ancient professional books on spleen were collected to be the analyzing source data.Using the Chinese medicinal herbs contained in the prescription as the object of study,then building the database of medicinal herbs related to spleen deficiency.The names of medicinal herbs were standardized according to the textbook of “Chinese Medicine”(the 7th ed.), Chinese Pharmacopoeia, and Chinese Materia Medica to make synonym and byname standard.The Association Rule analysis in the SPSS Clementine 12.0 was adopted to investigate the key medicinal herbs, the couplet medicines and the compatibility regularity in the prescriptions related to spleen deficiency.Results:1. 4422 research articles in CBM on spleen deficiency published in 1978-2014 were obtained and screened to be the analysis source data. The 106 diseases in traditional Chinese medicine are highly related to spleen deficiency like diarrhea, anorexia, constipation, metrorrhagia,diabetes, asthma, epigastric pain, phlegm and retained fluid, impairment by overeating; and278 diseases like diarrhea, ulcerative colitis, diabetes mellitus, dyspepsia, chronic hepatitis B,chronic renat farlure, irritable bowel syndrome, atrophic gastritis, constipation in modern medicine were found out according to the frequency of subject headings.2. 549 prescriptions related to spleen were obtained from China Prescription Database,involving 424 types of medicinal herbs, appeared 5194 times. The top 10 are Atractylodes,Poria, dried tangerine peel, ginseng, licorice, angelica, Radix Aucklandiae, yam, Radix Paeoniae Alba and Magnolia officinalis. 69 prescriptions on spleen deficiency treatment were screened from the spleen professional books, involving 128 kinds of medicinal herbs,appeared 614 times. The top 10 includ licorice, ginseng, dried tangerine peel, Atractylodes,Astragalus, poria, rhizome zingiberis, bupleurum, and Radix Paeoniae Alba.3. By association rules analysis, the common used 2 drugs combinations are Atractylodesporia, Atractylodes―dried tangerine peel, poria―ginseng, poria―dried tangerine peel, and Atractylodes―licorice. The common used 3 drugs combination are Atractylodes―poria―licorice, Atractylodes―dried tangerine peel―licorice, Atractylodes―poria―dried tangerine peel, Atractylodes―poria―ginseng, and poria―dried tangerine peel―licorice. The 4 drugs combination are Atractylodes―poria―dried tangerine peel―licorice, Atractylodes―poria―ginseng―licorice, Atractylodes―poria―dried tangerine peel―ginseng, Atractylodes―dried tangerine peel―ginseng―licorice, and poria―dried tangerine peel―ginseng―licorice.Conclusion:1. In traditional Chinese medicine, the diseases related to spleen deficiency are mainly in the spleen and stomach disorders, such as diarrhea, anorexia, constipation, metrorrhagia, diabetes,asthma, and epigastric pain. In modern medicine, the diseases related to spleen deficiency are mainly in gastrointestinal diseases, such as diarrhea, gastritis, ulcerative colitis, diabetes mellitus, dyspepsia, and chronic hepatitis B. That means the above diseases have a close relationship with spleen deficiency syndrome.2. The pattern of syndrome in traditional China medicine was insufficiency of the spleen-qi and deficiency of the spleen-yin.3. The medicinal herbs in the prescriptions are mainly for restoring vital energy and qi-regulating drugs. The subordinate are interior-warming drugs. The key drugs are Atractylodes, poria, dried tangerine peel, ginseng, licorice, angilica, and Radix Paeoniae Alba.4. The tropism of taste of medicinal herbs in the related prescription are mainly spleen and stomach meridian, Xin, Gan, and warm. Compatibility relationships in the prescriptions are mainly in Mutual promotion, and then mutual-assistance. As for the drugs combination, the effectiveness is mainly in the tonifying spleen and removing dampness, promoting circulaton of qi, supplementing qi, promoting digestion, and nourishing vitality and haemogenesis.5. The highly frequent used medicinal herbs can form some basic prescriptions for tonifying spleen, such as “Danggui Buxue Tang”,“Sijunzi Tang”, and “Yi Gong San”.
