Expression Of ZEB1 And Vimentin In Adenocarcinoma Of The Uterine Cervix(AUC)and Their Signigicances

Objective: To observe the expression of ZEB1(zinc-finger E-box binding homeobox1,ZEB1)and Vimentin in the tissue of normal cervical(NC),cervical adenocarcinoma in situ(AIS),adenocarcinoma of the uterine cervix(AUC),squamous carcinoma of the cervix(SCC),adenosquamous carcinoma of cervix(ASCC).To analyse the protein expression of ZEB1 and Vimentin,and to expound the relationship between their expression and the clinicopathological parameters.Evaluate the expression of two proteins' diagnosis value of different pathological types of cervical cancer.Methods :Applying Immunohistochemistry(IHC)Envision to detect expression of ZEB1 and Vimentin in each group.There are 66 cases in AUC group,12 cases in AIS group,15 cases in normal cervix group,45 cases in SCC group and 8 cases in ASCC group.SPSS20.0 is used to describe and deduct the statistics.The qualitative data is presented by frequency and percentage,the comparison between groups use Chi-square test or Fisher's exact test,linear trend date use linear trend Chi-square test.The area under the curve(AUC*)of ROC is used to compare diagnosis value under different different index of different hisopathological types.Spearman rank correlation is used to describe the correlation between two qualitative data.Significant level ?=0.05.When P <0.05,the difference is statistically significant.Results: 1.The positive expression rate of ZEB1 in NC group is 6.67%,25.00% in AIS group,75.76% in AUC group,and the positive expression rate is getting higher step by step.The comparisons among 3 groups are statistically significant(? 2=8.668,P=0.013).The comparisons between AUC group to NC group,AIS group are statistically significant(P<0.05),but it does not statistically significant(P>0.05)while take the NC group and AIS group in comparison.The positive expression is 57.78% in SCC group and 87.50% in ASCC group,the comparison in AUC group,SCC group and ASCC group is not statistically significant(? 2=5.463,P=0.065).2.The expression of ZEB1 is related to the patient's tumor invasive depth,FIGO period,histological differentiation extent,lymphatic metastasis,cancer embolus in vessels and subtypes of adenocarcinoma(P<0.05),it is not related to the size of tumor(P>0.05).3.The Vimentin protein did not express in NC group,while the positive expression rate of Vimentin in AIS is 16.67%,27.27% in AUC,the comparisons between 3 groups are statistically significant(? 2=8.668,P=0.013).Only the comparison between AIS group and NC group is statistically significant(P<0.05).Nither the AUC group to AIS group nor the AUC group to NC group are statistically significant(P>0.05).The expression of Vimentin is 11.11% in SCC,75% in ASCC.The comparisons between AUC group,SCC group and ASCC group are statistically significant(? 2 =14.424,P=0.001).The comparison between AUC group and SCC group,ASCC group is statistically significant(P<0.05).4.The presentation of Vimentin is related to the patient's FIGO period,histological differentiation extent,lymphatic metastasis,cancer embolus in vessels(P<0.05),it is not related to the size of tumor,tumor invasive depth,subtypes of adenocarcinoma(P>0.05).5.In cervical adenocarcinoma,the presentation of ZEB1 and Vimentin has positive correlation and is statistically significant.(r=0.325,P<0.001).Conclusion:1.The expression of ZEB1 and Vimentin are gradually elevated in the forming process of cervical adenocarcinoma.It speculates that ZEB1 and Vimentin may related to the development of cervical adenocarcinoma.2.The expression of ZEB1 and Vimentin are increased with the higher FIGO period and the decrease of tissue differentiation level;we speculate that ZEB1 and Vimentin may related to the grade malignancy of cervical adenocarcinoma.ZEB1 and Vimentin are represented high presentation in lymphatic metaphatic metastasis group and vascular invasion group.We surmise that ZEB1 and Vimentin may related to the invasion and metastasis of AUC.3.There is positive correlation between the expression of ZEB1 and Vimentin in AUC.The result suggests that the two proteins in the initiation and progression of AUC may have synergistic effect.
