Objective: to further study whether or not the acupuncture of Gallbladder Meridian of foot-Shaoyang has a similar regulatory effect on the weight of normal rat based on the previous confirmation that electric acupuncture of acupoint and Meridian of foot-Shaoyang can induce a reduction of weight in castrated osteoporosis rats and rats with type-II diabetes,and to explore the possible mechanism of acupuncture of Gallbladder Meridian of foot-Shaoyang in controlling the rat weight through a measurement of rat's food intake,weight,Lee's index and serum leptin,osteocalcin and ACTH.Methods : 20 clean-level SD male rats and 20 female ones were provided by Chongqing Tengxin experimental animal center,weighing 180-250 g.firstly,the experimental animals were raised for a week to get accustomed to the environment,and they were raised in a clean-level animal laboratory with a temperature of 22-26 ? and relative humidity of 60-80 %,fed by ordinary fodder,the rat whose weight was with ±15g was chosen and included into the experiment in a week.According to the principle of randomized controlled experiment,5 male and 5 female rats were randomly selected with a random number method and included into the electric acupuncture Gallbladder Meridian group and 5 male and 5 female rats were included into the control group.Followed by an electric acupuncture intervention for 4 weeksconsecutively.The meridians and acupoints intervened: Yanglingquan,Huantiao and Xuanzhong;control group: tied up in the same way as the electric acupuncture group without acupuncture intervention.The rats were recorded in terms of weight,amount of consumption of water and food during the first three weeks of experiment.In the 4th week,the abovementioned data was detected and measured every day.The body length of rats was measured before and after experiment for calculation of Lee's index.The rats were killed 28 days after intervention,and they underwent a food fasting for 12 hours before they were killed without water fasting,and then their weight was measured by an electronic platform scale,followed by an anesthesia through intraperitoneal injection of 2% pentobarbital sodium by 0.2-0.3ml/100 g of weight,then the animals were performed with a laparotomy to draw 10 ml of blood from the abdominal aorta after they were anesthetized,then the blood was placed into a refrigerator under 4? for 24 h,the serum was isolated(2000 rotation/min,10min)and placed into a cryogenic refrigerator for storage under-80?,awaiting a measurement of various biochemical indexes.Results: General condition and behavioral condition: before experiment,the female and male rats in electric acupuncture group were respectively compared with the ones in control group,the differences in body weight,consumption of food,drink and water,rat hair luster and color and activity were all insignificant.According to statistical analysis,the differences in pre-experiment body weight and consumption of food and drink between the male rats in electric acupuncture group and themale ones in control group were not statistically significant.The differences in pre-experiment body weight and consumption of food and drink between the female rats in electric acupuncture group and the female ones in control group were not statistically significant.1.Change in body weight: 1 week after experiment,the body weight of male rats in the electric acupuncture group became less than that of male rats in control group,the difference was statistically significant(P<0.05);the body weight of female rats in the electric acupuncture group became somewhat less than that of female rats in control group,but the difference was not statistically significant;from the 2nd to 4th week after experiment,the body weight of male rats in the electric acupuncture group became less than that of male rats in control group,the difference was statistically significant(P<0.05);the body weight of female rats in the electric acupuncture group became less than that of female rats in control group,the difference was statistically significant(P<0.05);4 weeks after experiment,the body weight of male rats in the electric acupuncture group became more than their pre-experiment body weight to some extent,the difference was statistically significant(P<0.05);but their body weight became significantly less than the body weight of male rats in control group,the difference was statistically significant(P<0.05);the body weight of female rats in the electric acupuncture group became more than their pre-experiment body weight to some extent,the difference was not statistically significant.but their body weight became significantly less than the body weight of female rats in controlgroup,the difference was statistically significant(P<0.05)? 2.Change in consumption of food and drink: in the 1st and 2nd week of experiment,the consumption of food and drink of rats in male electric acupuncture group became lower than that of rats in male control group,the difference between both was statistically significant(P<0.05),in the 3rd and 4th week,the difference between both was not statistically significant.Throughout the process of experiment,the difference in consumption of food and drink between the female electric acupuncture group and female control group was little and not statistically significant.3.Change in consumption of food and drink:in the 1st and 2nd week of experiment,the consumption of food and drink of rats in male electric acupuncture group became lower than that of rats in male control group,the difference between both was statistically significant(P<0.05),in the 3rd and 4th week,the difference between both was not statistically significant.Throughout the process of experiment,the difference in consumption of food and drink between the female electric acupuncture group and female control group was little and not statistically significant.The Lee's index of female rats in electric acupuncture group became lower than that of the female rats in control group,the difference between both was statistically significant(P<0.05).As for the male rats in electric acupuncture group,their Lee's index after experiment became lower than the one before experiment,the difference between both was statistically significant(P<0.05).As for the female rats in electric acupuncture group,their Lee's index after experiment becamelower than the one before experiment,the difference between both was statistically significant(P<0.05).As for the male rats in control group,the difference between pre-experiment and post-experiment Lee's index was little.As for the female rats in control group,the difference between pre-experiment Lee ' s index and post-experiment one was little.3.adrenocorticotrophic hormone(ACTH)content: the statistical mean numbers of ACTH of electric acupuncture group were all significantly lower than that of control group,but the difference in ACTH between the female electric acupuncture group and female control group was not statistically significant(P=0.078).The difference in ACTH between the male electric acupuncture group and male control group was statistically significant(P<0.05).4.Content of serum bone gla protein(BGP):the content of BGP of male electric acupuncture group was hardly different from that of the male control group,the difference was not statistically significant.The content of BGP of female electric acupuncture group was significantly different from that of the female control group,the difference between both was statistically significant(P<0.05).The difference in BGP content between male control group and female control group was nearly zero,which was not statistically significant.The difference in BGP content between male control group and female control group was significant,which was statistically significant.(P<0.05).5.Content of serum bone gla protein(BGP):the content of BGP of male electric acupuncture group was hardly different from that of the male control group,the difference was not statisticallysignificant.The content of BGP of female electric acupuncture group was significantly different from that of the female control group,the difference between both was statistically significant(P<0.05).The difference in BGP content between male control group and female control group was nearly zero,which was not statistically significant.The difference in BGP content between male control group and female control group was significant,which was statistically significant.(P<0.05)Conclusion: 1.the electric acupuncture of Gallbladder Meridian of foot-Shaoyang has an effect of inhibiting the growth of body weight on the normal rats.2.the mechanism of physiological weight loss of rat caused by the electric acupuncture of Gallbladder Meridian of foot-Shaoyang has nothing to do with the direct inhibition of appetite.3 the electric acupuncture of Gallbladder Meridian of foot-Shaoyang causes a reduction of ACTH in physiological rats,where it has a significant effect of reducing ACTH in physiological male rats while tends to reduce the ACTH in female rats,and it can also lead to a significant reduction of BGP in physiological female rats,while its effect on the BGP of male rats is insignificant,all of which suggest that,the weight loss of physiological rats may be associated with the decrease in content of ACTH and BGP in serum.4.Conclusion: the electric acupuncture of Gallbladder Meridian of foot-Shaoyang doesn't have a specific effect on the content of leptin of normal rat.
