Objective: To observe changes of stomach and gallbladder movement and the related brain-gut peptide(BGP) receptor genes expression by Electroacupuncture (EA) in Acupoints of Foot-Yangming meridian and Foot-Shaoyang meridian respectively. Methods: Electromyography of Rabbits' stomachs and Oddi sphincters were recorded respectively by double-pathway physiological grapher when the animal were in movement-inhibited status with EA in Sibai and Zusanli of the Foot-Yangming meridian, Yanglingquan of the Foot-Shaoyang meridian, and Chengjin of the Foot-Taiyang meridian. Motilin (MTL) of serum, the gastric antrum and Oddi sphincter, cholecystokinin (CCK) were tested by radioimmuno- assay (RIA). And MTL mRNA and CCKA-R mRNA were carried out by RT-PCR.Results: 1. The high movement phase of slow wave and the amplitude of fast wave in electromyography of Rabbits' stomachs and Oddi sphincters respectively showed excited reaction with different levels when EA in Sibai, Zusanli and Yanglingquan with significant difference (p<0.05 or 0.01), compared with Atropine group and Chengjin group. 2. MTL of serum, the gastric antrum'ssmooth muscle and Oddi sphincter, as well as CCK quantity increased when EA in Sibai, Zusanli and Yanglingquan with significant difference (p<0.05 or 0.01), compared with Atropine group and Chengjin group. 3. MTL mRNA of the gastric antrum's smooth muscle and Oddi sphincter, as well as CCKA-R mRNA expression increased massively when EA in Sibai, Zusanli and Yanglingquan with significant difference (p<0.05 or 0.01), compared with Atropine group and Chengjin group. 4. EA in Sibai could get a synchronization with variety of electromyography of the stomach and Oddi sphincter, release of MTL and CCK, expression of MTL mRNA and CCKA-R mRNA, which were better than the effect of Zusanli and Yanglingquan could do respectively. Conclusions: 1.There is a relative particularity between Foot-Yangming meridian and stomach, so do Foot-Shaoyang meridian and gallbladder, which are embodied on regulating stomach and gallbladder functions respectively. 2. This research has further improved that the peptides are the material of Meridians and Viscus Theory, and acupuncture efficacy would come true by different BGPs release and regulation. 3. EA in acupoints of Foot-Yangming meridian and Foot-Shaoyang meridian respectively could make MTL mRNA and CCKA-R mRNA expression up-regulation which shows that acupuncture efficacy are encompassed with the molecular level changes. 4.The mechanism of EA in Sibai which would regulate significantly the function of the stomach and gallbladder is still deserved deep research.
