Study On The LTBP1 Of Kidney Yang Deficiency Syndrome Of TCM In Postmenopausal Women

Objective:To study the correlation between kidney yang deficiency and LTBP1 in postmenopausal osteoporosis,and to provide reference for the study of the syndrome of kidney yang deficiency syndrome.Methods:Select the postmenopausal women in Fuzhou area,through questionnaire survey,dual energy X-ray bone mineral density detection and TCM syndrome differentiation,divided into PMOP kidney yin deficiency group(32 cases),PMOP kidney yang deficiency group(n = 25),control group(n = 32).The venous blood was collected and the white blood cells were separated.The total RNA was extracted by one step method of Trizol,and the quality of RNA was detected by RNA.The sample was cDNA.The purpose of each gene(LTBP1)and reference gene(GAPDH)of the RT-PCR reaction,draw the gradient dilution DNA standard curve,the concentration of each sample divided by LTBP1 GAPDH,the relative concentration of the samples after correction of gene.PMOP technique was used to detect the expression of LTBP1 protein in the kidney yang deficiency syndrome group(25 cases)and the control group(n = 25)by Western-blot.Finally,SPSS20.0 statistics,comparing the expression level of LTBP1 in each group.Results:?No significant differences were age,menarche age,menopausal age,BMI,the relative concentration of LTBP1 after correction were:PMOP kidney yin deficiency group:0.055 +0.060 PMOP;kidney yang deficiency group:0.010 ± 0.011;control group:0.027 ±0.026;? PMOP in kidney yang deficiency group compared with control group,PMOP of kidney yang deficiency group and control group LTBP1 mRNA expression level difference(P=0.004),and the expression level is lower than that of the control group;? PMOP kidney yin deficiency group and control group LTBP1 mRNA expression level was not statistically significant(P=0.099);?There was a statistically significant difference between the expression level of PMOP and PMOP of kidney yang deficiency of kidney yin deficiency group.Group LTBP1(mRNA P=0.001);?The control group:the gray value of LTBP1 0.687± 0.592 PMOP,kidney yang syndrome group:0.362 ± 0.289 PMOP,and the kidney yang deficiency group Compared with group PMOP,the expression level of LTBP1 protein in kidney yang deficiency syndrome group was significantly lower than that in control group(Wilcoxon rank sum test,Z=-2.202,P=0.028),the difference was statistically significant..Conclusions:Compared with the PMOP of kidney yang deficiency group and control group,the downregulation of LTBP1(P<O.01),the detection and gene chip in the previous study results are the same,further confirmed:osteoporosis of kidney yang deficiency and the expression of LTBP1 between postmenopausal women.
