| Objective: To investigate the correlation between Klotho gene polymorphism a nd serum and urine biochemical index and the risk of idiopathic calcium oxalate stones(ICOS)in Uygur population in Xinjiang.Method: A total of 128 Uygur idiopathic calcium oxalate kidney stones(ICOS group)and 94 normal subjects(control group)were collected.Peripheral venous blood was collected and genomic DNA was extracted.Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)technique was used to detect the rs3752472 polymorphism locus of Klotho gene in the two groups.The serum biochemical and urine samples were collected and analyzed.The distribution of genotype in ICOS group and control group was compared and analyzed Differences in blood and urine biochemical markers in genotype carriers.Results: The genotype and allele frequencies of the Klotho polymorphism loci rs3752472 were consistent with the Hardy-Weinberg equilibrium law.The ICOS group compared with the control group,the risk of calcium oxalate was higher in the Kloto rs3752472 locus CC homozygous genotype carriers(OR=3.707,95%CI=1.8617.384,P=0.0001).Compared with the T allele carrie-rs,the C allele carriers were significantly higher than those of the T allele carrier-s(OR=3.675,95%CI=1.9769.838,P=0.0001);The levels of potassium,sodium,calcium,phosphor us and carbon dioxide(CO2CP),triglyceride(TG)and hi-gh density lipoprotein(HDL)and urine were significantly different between the IC-OS group and the contro l group in the blood and urine biochemical observation i-ndexes(P<0.05).The rel ationship between the genotype of Klotho rs3752472 pol-ymorphism and blood and urine biochemical indexes was found to be significant(P<0.05),and the difference was statistically significant(P<0.05).The levels of serum calcium,phosphorus and TG in CC genotype carriers were significantly hig-her than those in CC + TT genotype(P<0.05)Calcium CC genotype carriers w-ere higher than CC + TT gen otype carriers,the difference was statistically significa-nt(P<0.05).Conclusions: The study population was significantly related to Klot-ho gene rs3752472 polymorphi sm and pathogenesis,which may be an independent risk factor for the onset of ICOS;it may be based on serum electrolytes(calcium,phosphorus)and lipids(TG)pathogenesis level involved in regulation of ICOS. |
