Objective:1、To investigate the expression and the interrelation of osteopontin(OPN) and CD44 in the renal tissues of patients which have idiopathic calcium oxalate stone (ICS);2、To investigate the relationship between OPN, CD44 and the formation of ICS which provide reference and ideas for the further study of the ICS pathogenesis.Methods:Selected 57 cases renal tissues with ICS induced renal non-functional for surgical resection as the experimental group from January 2011 to June 2012 in Urology department of the first Affiliated Hospital of Gannan Medical University,at the same time,selected 30 cases renal tissues beside tumor specimens (radical nephrectomy patients were confirmed by pathology) as the control group.The expression of OPN and CD44 in the renal tissues of patients with ICS were tested by immunohistochemical method(SP); The expression of OPN mRNA in the renal tissues of patients with ICS were tested by reverse transcription-polymerase chain reaction method(RT-PCR), And to compare the two sets of results between the immunohistochemical method and reverse transcription-polymerase chain reaction method.Results:1、The positive expression rate of OPN and CD44 protein were 57.9%(33/57) and 54.4%(31/57) in the experimental group, but in the control group were 16.7% (5/30) and 10.0%(3/30), the difference between the two groups were statistically significant (P<0.05). The OPN and the CD44 protein expression in the experimental group were positively correlated (Kendall’s tau-b=0.503, P<0.05).2、45 samples amplified OPN specific bands in the experimental group, the positive rate was 78.9%, the relative content of OPN mRNA was 0.571±0.106. There were 7 samples can amplify OPN specific bands in the control group, the positive rate was 23.3% and the OPN.mRNA relative content was 0.356±0.106. The difference between the two groups were statistically significant (P<0.05).Conclusions:1、The expression of OPN and its receptor CD44 are correlation with the formation of the ICS, and may promote the formation of the ICS;2、OPN may promote the formation of the ICS through its receptor CD44;3、The expression of OPN may be affected by some external or endogenous factors.
