Objective: To investigate the correlation between multiple sites of Klotho gene rs1207568,rs3752472,rs564481,rs648202 and rs650439 and Uygur idiopathic ca lcium oxalate kidney stones in Xinjiang.Methods: A total of 400 Uygur patients with calcium oxalate kidney stones and 410 healthy subjects were selected.The Klo-tho gene polymophisms rs1207568,rs3752472,rs564481,rs648202 and rs650439 genotypes and alleles were determined by PCR and DNA sequencing.The frequency of gene distribution,and the correlation between the relevant gene locus and the onset of idiopathic calcium oxalate kidney stones and the effects on blood lipids and blood biochemistry.Results: The genotype distribution of the selected Uyghur case group and the normal group was consistent with the Hardy-Weinberg equilibrium,and the selected study population was representative.The risk of allele C was higher in the Klothors1207568 case group than in the control group(OR=1.295,95%CI: 1.019~1.645,P=0.034).Residual Klotho gene rs3752472(OR = 1.266,95%CI:0.861~1.858,P = 0.226),rs564481(OR = 1.120,95%CI: 0.906~1.384,P = 0.295),rs648202(OR = 0.916,95%CI:0.732~1.141,P = 0.435),rs650439(OR=0.994,95%CI:0.794~1.239,P=0.960)There was no significant difference in allele distribution(P>0.05).Compared with the CT+TT genotype in the Klotho gene rs1207568 locus case,blood calcium(t=3.421,P=0.001),blood phosphorus(t=2.952,P=0.004),blood chlorine(t=4.076,P=0.001),urea nitrogen(t=2.925,P=0.004),uric acid(t=3.614,P=0.001)were higher than the latter,with statistically significant(P<0.01),and magnesium.(t=1.086,P=0.279),serum potassium(t=0.446,P=0.656),blood sodium level(t=0.146,P=0.884)were not statistically significant;blood lipids were TG(t=2.259,P<0.026),TC(t=2.319,P<0.022)were statistically significant,while HDL-C(t=0.736,P=0.447)and LDL-C(t=1.659,P=0.100)were com-pared.Statistical significance(P > 0.01).Conclusion: The Klotho gene rs3752472,rs564481,rs648202 and rs650439 have no significant correlation with the incidence of calcium oxalate kidney stones in Xinjiang Uygur population,so it may not be a risk factor for calcium oxalate stone.There is a significant correlation between the incidence of Klotho gene rs1207568,and the C allele is one of the risk factors for idiopathic calcium oxalate kidney stones.The effect of Klotho gene rs1207568 on calcium oxalate kidney stones may affect the formation of calcium oxalate stones by affecting calcium and phosphorus metabolism,blood lipids TG and TC.
