Chapter 1: Correlation between the polymorphisms of Klotho gene and renal calcium oxalate stones susceptibility in the Uygur nationality: A case-control studyObjective: To investigate the correlation between gene polymorphisms of Klotho gene and renal calcium oxalate stones susceptibility in the Uygur nationality. M ethod: 129 patients with calciu-m oxalate stones(case group) and 94 healthy people(control group) were enrolled in our study, peripheral blood was collected and genomic DNA were extracted from peripheral blood, polymerase chain reaction restriction fragment length polymorphism(PCR-RFLP) was performed to analysis polymorphisms rs3752472, rs650439 and rs1207568 of Klotho, compare association between different genetypes and calcium oxalate stones susceptibility, and made a comprehensive analysis of these SNP locies distribution of alleles in Xinjiang Uygur calcium oxalate ston-es patients and healthy people. Results: The results of genetype and alleles of Klotho gene reveals that the distributation of three locis goes with Hardy-Weinberg equilibrium. For Klotho gene rs3752472 site, the statistical results of its genotype and allelefrequencies have statistical significance between the two groups(P<0.05). The other two gene sites have not statistical significance between the two groups(P>0.05).Conclusions: The rs3752472 polymorphism of K lotho gene is associated with the risk of calcium oxalate urolithiasis in Xin Jiang region, and may be one of risk factors in the pathogenesis of calcium oxalate stones. rs650439 and rs1207568 polymorphisms of Klotho gene has no stasitically association with calcium oxalate stones susceptibility. Klotho gene can inhibit the formation of calcium oxalate stones by some unfound molecular mechanisms, our results may still need further study.Chapter 2: Correlation between Klotho gene and protein and SD rats with calcium oxalate nephrolithiasisObjective: To investigate the correlation between Klotho gene and renal calcium oxalate stones in SD rats. Methods: 15 SD rats in each were chosen in the e xperimental group and control group to study Klotho and TRPV5 gene, 18 SD rats in the experimental group and 9 SD rats in control group were chosen to study the Klotho and TRPV5 protein. In the SPF laboratory every day each rat in the experimental group was fed with 2% 2ml ammonium chloride soluteion and 1% glycol solution free drinking water. The control group was given normal water. Other environment and conditions are equal. All the SD rats were fed for four weeks. Kl otho gene and TRPV5 gene in kidney tissue were detected in the two groups by f luorescence quantitative PCR technology and Klotho protein and TRPV5 protein in kidney tissue were detected in the two groups by Western Blot technology.Results:Klotho gene and its protein and TRPV5 gene and its protein in the experimental g roup were lower than that in the control group and they had statistical significance(P<0.05). Conclusion: Klotho gene may be the formation of calcium oxalate stones susceptibility gene and can inhibit the formation of calcium oxalate stones by stimulating the TRPV5 expression, but whether it is the susceptibility gene of human calcium oxalate stones and the specific mechanism of action still need further r esearch.
