Objective:To.explore the association of urokinase gene3′-UTR T/C polymorph-hism with idiopathic calcium oxalate urolithiasis in Han population in JiangxiProvince.Methods:In this study,80healthy people having no history of stone formation,and87people who had idiopathic calcium oxalate stones were enrolled. Urokinase gene3′-UTR T/C polymorphism was determined by polymerase chain reaction andrestriction fraction length polymorphism(PCR-RFLP).Result:Differences of age,sex composition and body mass index(BMI) between thetwo groups were not significant. The distribution frequencies of Urokinase gene3′-UTR T/C polymorphism genotype and alleles in the two groups followed theHardy-Weinberg equilibrium(χ~2=3.289,P=0.07;χ~2=3.097,P=0.078,P>0.05). Thedifference of frequencies of the TT,CT+CC genotypes between the two groups weresignificant[χ~2=4.445,P=0.035<0.05,OR=1.934,95%CI(1.045,3.582)]. Thedifference of frequencies of the T and C alleles between the two groups were alsosignificant[χ~2=6.330,P=0.012<0.05,OR=1.796,95%CI(1.136,2.840)].Conclusions:1. There is significant association between urokinase gene3′-UTR T/Cpolymorphism with idiopathic calcium oxalate urolithiasis.2. The TT genotype may be a good genetic marker for idiopathic calcium oxalatenephrolithiasis. The subjects with TT genotype yielded a1.934fold increased riskfor calcium oxalate nephrolithiasis.3. Allele T is an risk factor for idiopathic calcium oxalate nephrolithiasis. Thesubjects with T yielded a1.934fold increased risk for idiopathic calcium oxalatenephrolithiasis. |
