ObjectiveTo investigate the correlation between MitoKATP and mechanism of Z hishi Xiebai Guizhi Decotion in protection of myocardial ischemia reper fusion injury rabbtis by using Langendorff isolated heart perfusion syste rm.MethodsUsing the BL-420F biological function experimental system to detect the II lead of ECG in healthy adult rabbits,and to remove the abnorm al ECG,and to replicate the rabbit model of Langendorff perfusion in vitro.The 40 rabbits were randomly divided into 5 groups(n=8):non-ischemia reperfusion group(sham group),ischemia reperfusion group(M IRI group),ischemia reperfusion+Zhishi Xiebai Guizhi Decoction group(MIRI+ZSXBGZD group),ischemia reperfusion+Zhishi Xiebai Guizhi De coction+5HD group(5-HDgroup),ischemia,reperfusion+Zhishi Xiebai Gui zhi Decoction+glibencLamide group(Gli group);MIRI model was made t o observe the changes of The heart hemodynamic of rabbits,including L eft ventricular systolic pressure(LVSP),left ventricular end diastolic pr essure(LVEDP),left ventricular pressure maximum the rise and decline rate(ądP/dt max)effect,and to measure the physiological and pathol ogical indexes of SOD,MDA and LDH,the myocardial infarct size dye d by TTC methods,and the coronary flow rate(CF)in isolated rabbits with MIRI.Results1 All the indexes were up to the mustard of the experiments:the left ventricular systolic pressure(LVSP),the left ventricular end diastolic pressure(LVEDP),the maximum rise and fall rate of left ventricular pr essure(ądP/dt max)and coronary flow(CF)were recorded successfully.The whole experimental process was stable,and the beating time of ra bbit heart in vitro was long.2 Through the analysis of the isolated rabbit heart left ventricular he modynamic curve and as compared with Sham group,LVEDP in MIRI gr oup was significantly increased,LVSP5,ądP/dt max and CF in MIRI gro up were decreased,existing a a significant difference;as compared with MIRI group,:LVEDP in MIRI+ZSXBGZD group was decreased,LVSP,ądP/dt max and CF were increased,embraving a significant difference.3 by the analysis of pathological indexes of MIRI group,MDA of m yocardium tissue was significantly higher than that of Sham group P<0.01,SOD activity of myocardial tissue was significantly lower than that of Sham group P<0.01;myocardial tissue of rabbits in MIRI+ZSXBGZ D group were significantly lower than MIRI group MDA P<0.01,SOD activity of myocardial tissue was significantly higher than that of MIRI group P<0.01,there were significant differences.4 Myocardial infarction area in MIRI group were significantly higher than that in Sham group,there were statistically significant differences(P<0.05);compared with MIRI group,Myocardial infarct size reduce d in ZSXBGZD group,the differences were significant(P<0.05).Com pared with MIRI group,there was no significant change in myocardial i nfarction area between the 5-HD group and Gli group(P>0,05),but si gnificant difference was existed between MIRI group and ZSXBGZD gro up.Conclusion1 The MIRI model was successfully established and the stable hemod ynamic curve was recorded,which showed that LVSP decreased,LVEDP increased,and ądP/dt/max slowed down.2 Zhishi Xiebai Guizhi Decoction can improve the metabolic level of myocardial tissue perfusion of rabbits with MIRI,and increase corona ry flow.3 Zhishi Xiebai Guizhi Decoction can enhance the activity of SOD,reduce the content of MDA,LDH by scavenging oxygen free radicals,a gainst lipid peroxidation to alleviate myocardial ischemia reperfusion inj ury.4 Zhishi Xiebai Guizhi Decoction can take an advantages of reducin g myocardial infarct size of rabbits with MIRI,and improving heart fun ction.5 Zhishi Xiebai Guizhi Decotion can reduce injury of MIRI in rabbi ts and protect myocardium tissue,which can be largely offset by 5-H D and can be completely blocked by Gli,showed that the protective effe cts of Zhishi Xiebai Guizhi Decotion on MIRI rabbits were significantl y associated with MitoKATP.
