Objective:Observe the protective effect of Zhishi Xiebai Guizhi Decoction on ET-1 induced acute myocardial ischemia in rabbits,For clinical safety,scientific and reasonable to use this medicine to provide experimental data for the treatment of ischemic heart disease.Methods:60 healthy adult rabbits,Including male and female,Randomly divided into 6 groups(10/group),which were Control group,model group,Zhishi Xiebai Guizhi Decoction of high,medium and low dose group,positive drug group(Danshen Pill).Control group and model group were given equal volume of normal saline(supplementary).(experimental animals were provided by the experimental animal center of Heilongjiang University Of Chinese Medicine).According to the MR.Wu's method,the rabbits were modeled in gavage after fifth days.First of all,to record the limb lead ECG,electrocardiogram is not removed in normal rabbits,except the control group,other groups were through ear vein bolus injection(rapid intravenous drug injection of small dose of 1.4nmol/kg.ET-1).ET-1 Induced Acute Myocardial Ischemia in rabbits.We recorded 10s?1min?5min?10min?30min?60min?120min limb,electrocardiogram and ventricular pressure.After 2h,the orbital venous blood,serum separation,stored in the-20 C refrigerator,CK-MB,measured CK,LDH,MDA,sod.Then,the rabbits were killed immediately.We take the pathology after heart for determination of moisture content,drying oven at 60 DEG C 24h,measured dry weight.We calculate the myocardial tissue water content.Result:1.Rabbits in the model group of ECG HR decreased significantly,T wave tip,ST segment change obviously,compared with the control electrocardiogram of rabbits in the model group,there was significant difference.LVSP in the model group decreased,LVEDP increased,?dp/dtmax decreased.Compared with the control group,there was significant difference.The model group serum LDH,CK,CK-MB increased significantly,MDA content increased,SOD activity decreased,compared with the control group,there was significant difference.The percentage of water content in model group increased compared with the control group,there was significant difference.2.Zhishi Xiebai Guizhi Decoction high,middle dose group and Model Group HR increased,St,T wave amplitude decreased,LVSP increased,the increase of dp/dtmax,LDH,CK,CK-MB decreased,MDA decreased,SOD increased,myocardial water content percentage decreased compared with the model group.There was significant difference between the low dose group compared with the model group,there was no significant difference.3?The positive control group compared with the model group,there were significant differences.Conclusion:1?This experiment through the ear vein bolus injectionET-1(1.4nmol/kg)can replicate the success of acute myocardial ischemia rats.2?Zhishi Xiebai Guizhi Decoction,high dose group of electrocardiogram in acute myocardial ischemia rabbits and ventricular pressure has obvious protective effects on ET-1 Induced Myocardial enzyme level,which fell back,enhance the antioxidant capacity,myocardial tissue water content decreased.3?Zhishi Xiebai Guizhi Decoction,high dose group of ET-1 Induced protective effect of acute myocardial ischemia model in rabbits,is the main mechanism of Zhishi Xiebai Guizhi Decoction can enhance the activity of free radical antioxidant enzyme system,the protection of the myocardial cell membrane,maintain cell membrane stability,improve ventricular pressure,promote the recovery of heart function.
