Objective:Induced by exogenous injection of ET-1 the rabbit model of coronary artery spasm,observe the blood vessel activity during contraction factor,such as oxidative stress mechanism in the role of vasospasm,compare the scallions frutus aurantii immaturus Bai Guizhi soup with shu to farmar,the intervention effect of diltiazem,and explore its mechanism.Methods:Choose healthy adult rabbits,a total of 48,randomly divided into 6 groups,model group,low dose,equivalent dose and high dose of acid-insoluble ash scallions Bai Guizhi soup group,diltiazem group,the method is easy to set,the experiment began to fill the stomach before 5,at the end of the time to fill the stomach after 1 hour,rabbit ear margin vein projectile injection ET-1,acute coronary spasm induced by myocardial ischemia model was established.Observe ecg indexes including the rabbit T wave height percentage,focal myocardial necrosis serum markers of myocardial calcium protein(cTn I)content,intravascular I original endothelin precursor BIGET-1 level,serum top ring element(PGI2),serum blood clots alkanes element level B2(TXB2),glutathione(GSH)level in serum,serum total superoxide dismutase(T SOD)activity and serum total antioxidant capacity(T AOC)and intravascular incidence during contraction factor NO level,serum malondialdehyde(MDA)concentration and myocardial pathologic histology changes.All data are expressed as a mean+standard deviation.Comparison between multiple sets of data using single factor analysis of variance(one-way ANOVA),data comparison in the group by t test.Results:1.By exogenous ET 1 May induce rabbit coronary artery spasm induced by myocardial ischemia,caused the T wave rise,focal myocardial necrosis markers elevated cTnl,myocardial structure is damaged,the focal necrosis,myocardial interstitial edema,accompanied by a small amount of inflammatory cells infiltration.2.Model group,the rabbit vascular intravascular incidence during contraction factor NO levels,elevated levels of endogenous endothelin precursor BIGET-1,there were NO changes in serum PGI2 levels,elevated serum TXB2 levels,serum GSH,T-SOD,T-AOC level decreased,MDA levels.3.Acid-insoluble ash,scallions Bai Guizhi soup equivalent dose and high dose and Rho kinase blocker shu to farmar,calcium channel blockers diltiazem can reduce T wave height,lower cTnI levels,reduce the focal point of necrosis.4.Acid-insoluble ash,scallions Bai Guizhi soup equivalent dose and high dose could increase the NO level,lower BIGET-1 level,increase the PGI2 levels,reduce TXB2 level,increase the GSH,T-SOD,T-AOC level,reduce the MDA level.5.Acid-insoluble ash scallions Bai Guizhi soup equivalent dose and high dose can obviously increase the PGI2 levels,and Rho kinase blocker shu to farmar,calcium channel blockers diltiazem exist significant differences.6.Acid-insoluble ash,scallions Bai Guizhi soup equivalent dose and high dose decrease serum TXB2 ability significant differences with diltiazem.7.Acid-insoluble ash,scallions Bai Guizhi soup equivalent dose and high dose in increasing serum T-AOC,and method of shu,and significant differences diltiazem.Conclusion:1.By exogenous ET 1 May induce rabbit coronary artery spasm caused by myocardial ischemia change.2.Exogenous ET 1 May induce rabbit coronary artery spasm caused by myocardial ischemia is associated with oxidative stress injury.3.Acid-insoluble ash,scallions Bai Guizhi soup equivalent dose and high dose and Rho kinase blocker shu to farmar,calcium channel blockers diltiazem can intervene by exogenous ET 1 May induce rabbit coronary artery spasm caused by changes of myocardial ischemia.4.Acid-insoluble ash,scallions Bai Guizhi soup equivalent dose and high dose of exogenous coronary artery spasm induced by injection of ET-1 the rabbit to the intervention of myocardial ischemia is associated with resistance to oxidative stress injury.5.Acid-insoluble ash scallions Bai Guizhi soup equivalent dose and high dose of exogenous induced by ET-1 rabbit coronary artery spasm induced by myocardial ischemia of intervention in the process,can significantly increase the PGI2 levels,and Rho kinase blocker shu to farmar,calcium channel blockers diltiazem has no this function.6.Acid-insoluble ash,scallions Bai Guizhi soup equivalent dose and high dose reduce exogenous induced by ET-1 rabbit coronary artery spasm induced by myocardial ischemia rabbit serum TXB2 ability obviously better than that of diltiazem.7.Acid-insoluble ash,scallions Bai Guizhi soup equivalent dose and high dose in improving exogenous induced by ET-1 rabbit coronary artery spasm induced by myocardial ischemia rabbit serum T-AOC,method is obviously better than the shu,and diltiazem.
