Objective:To study the Zhishi Xiebai Guizhi granules of anti myocardial ischemia effect and the possible mechanism, to provide the experimental basis for the research and development of new drugs and clinical application.Methods:the ECG screening qualified 72 rats, were randomly divided into normal group,model group, Zhishi Xiebai Guizhi granules in high dose group, middle dose group and low dose group, positive control group (Compound Salvia Tablet). Were orally given Zhishi Xiebai Guizhi granules (concentration of 1.2g/kg,0.6g/kg and 0.3g/kg) and Compound Salvia Tablet (concentration of 0.15g/kg) 1 times a day, for 7 consecutive days. Administration of the start of the fifth day, except the normal group, the other groups with isoproterenol (ISO) intraperitoneal injection (5mg/kg), for 3 consecutive days, preparationof acute myocardial ischemia rats. At the end of intraperitoneal injection of isoproterenol 6 hours later, the rats were examined ECG S-T segment changes;blood serum myocardial enzymes CK, LDH, AST activity, and the index of blood rheology; blood detecte d bySpectrophotometry in MDA, SOD, GSH-Px and NO levels; their hearts were detected b-yhistopathology; immunohistochemistry was used to detect the expression of Bax and Bcl-2 protein in myocardium.Results:1, ECG showed the rats in the model group of S-T segment elevation, there was significant difference compared with the normal group (p<0.05); the lower drug group S-T ratscompared with model group, in varying degrees, of which Zhishi Xie Bai Guizhi granules in high dose group showed the most obvious, compared with the model group were significantly the difference (p<0.05).2, compared with the normal group, the model group rats serum myocardial enzymes LDH,CK and AST activity increased significantly (p<0.05); all the rats in the treatment groupdecreased serum myocardial enzymes LDH, CK and AST activity in different degree, there is statistical significance compared with the model group (p<0.05); wherein the Zhishi Xiebai Guizhi granules with the dosage increased, the change of serum myocardial enzymes in rats with myocardial ischemia is the most obvious, in a dose dependent.3, compared with normal group, model group, the whole blood viscosity, plasma viscosityand fibrinogen were significantly increased (p<0.05); compared with the model group,Zhishi Xiebai Guizhi granules in high dose group, middle dose group and the positive control group, the index decreased significantly (p<0.05).4, compared with the normal group, model group, MDA content increased significantly,SOD, NO content decreased, GSH-Px activity decreased significantly, with statistical significance (p<0.05); compared with the model group, the treatment group of the aboveindexes were improved in different degrees, with statistical significance (P< 0.05).5, the pathological results showed:disordered fiber myocardial ischemia rats in the model group, partially ruptured, cytoplasm eosinophilic change obviously, the gap in the visiblelight to moderate hyperemia, edema and infiltration of inflammatory cells, nucleus concentrate is not clear, nucleolus. Compared with the model group, the treatment grouprats myocardial above pathological changes have different degree reduced, myocardial cells were arranged n neat, nuclear pyknosis decreased n number, acidophilic changereduce, reduce the number of inflammatory cells.6, compared with the normal group, model group of myocardial tissue expression level ofBax protein was significantly increased (p<0.05), the expression level of Bcl-2 protein was significantly decreased (p<0.05); compared with the model group, the expression level ofZhishi Xiebai Guizhi granules in high dose group, middle dose group, low dose group,positive medicine group decreased significantly (Bax protein p<0.05), the expression level of Bcl-2 protein was significantly increased (p<0.05).Conclusion:The pathological injury in l,Zhishi Xiebai Guizhi granules can be effective againstmyocardial ischemia induced by, has obvious protective effects on myocardial ischemia;Has obvious effect 2, Zhishi Xiebai Guizhi granules on rat myocardial ischemia ECG S-T segment change;3, Zhishi Xiebai Guizhi granules could obviously decrease the myocardial ischemia of ratsserum myocardial enzymes, LDH, CK, AST activity, can be reduced due to myocardial ischemia and hypoxia caused by the injury;4, Zhishi Xiebai Guizhi granules can make the myocardial ischemia of rats abnormally elevated whole blood viscosity, plasma viscosity and fibrinogen levels decreased significantly, improve blood viscosity concentration of condensed state;5, Zhishi Xiebai Guizhi granules probably by scavenging oxygen free radicals, improve the body’s antioxidant capacity, so as to improve the state of myocardial ischemia, plays the role of myocardial protection;6, Zhishi Xiebai Guizhi granules can make the protein down regulated expression of Bax in rats with myocardial ischemia, Bcl-2 protein expression level increased, thereby inhibitingthe apoptosis of myocardial cells.
