In this experiment, seventy five female ICR mice (about six weeks) were randomly divi-ded into 3 groups, namely control group, CTX group ? and CTX group ?, and were res-pectively given 0 mg/kg,0.6 mg/kg and 6 mg/kg of cyclophosphamide by intraperitoneal injection for 7 days. The blood samples were obtained by ophthalmectomy on the 1st? 4th?7th?10th?14th and 21st day of the experiment. WBC count and WBC differential count were detected by blood counter system. The percentages of B-lymphocyte and T-lym subsets in the peripheral blood were determined by flow cytometer. Using Th1/Th2/ Th17 kit, the serum cytokines were analyzed by Cytometric Bead Array, and the antioxidante nzyme activity and oxidation products in serum were detected by biochemistry kits. The results were as follows:CTX group ? The numbers of WBC and Lym in peripheral blood were increased at the 1st day, the number of Mon increased at the 1st and 4th day and the number of Gran increased at the 1st,4th and 10th day. The percentage of CD19+B-lym were decreased at 14th and 21st day. The percentage of CD3+ and CD3+CD4+T-lym were decreased at 7th day and increased at 14th and 21st day. In administration period, the contents of serum IL-2?IFN-??IL-4 and IL-10 were decreased, but the contents of TNF and IL-6 were increased. In withdrawl period, the contents of IFN-? and IL-4 were increased, the contents of TNF were decreased, and the contents of IL-2?IL-10 and IL-6 showed fluctuation. The contents of IL-17 were increased at the 1st and 4th day and showed fluctuation from 7th to 21st days. The activity of CAT and SOD, and the ability of inhibiting OH- were increased at the 1st,4th and 7th day. The contents of MDA were increased at 10th day.CTX group ? The umber of WBC and Lym in peripheral blood were decreased at the 4th and 7th day. The umber of WBC?Mon?Gran and Lym were increased at the 10th day. The percentage of CD19+B-lym were decreased in administration period and increased in withdrawl period. The percentage of CD3+ and CD3+CD4+T-lym were decreased at 7th day and 10th day, showed increase and decrease respectively at the 14th and 21st day. The percentage of CD3+CD8+T-lym were showed down-regulation tendency in total test period. The contents of serum IL-2, IFN-?, IL-4 and IL-10 were decreased in administration period and various increased in withdrawl period. The content of TNF was decreased at the 1st day, increased at the 7th and 10th day, and decreased at the 14th day. The content of IL-6 were decreased at the 1st to 4th days, increased at the 7th and 10th day, decreased at the 14th and 21st day. The content of IL-17 were decreased at the 4th, 7th, and 21st day. The activities of SOD and CAT were decreased, except that the activity of CAT was increased at the 4th day. The ability of inhibiting OH-were decreased, and the contents of MDA were increased.The above-mentioned results showed that 0.6mg/kg CTX might promote nonspecific immune function in mice, but the effects on different kind of white blood cells were obvious different.6 mg/kg CTX could induce immune suppression in administration period, but might facilitate immune function in withdrawl period.
