Effect Of Bushen Huiyang Recipe On Peripheral Blood Cytokines And Lymphocyte Subsets In Kidney Yang Deficiency Mice

Nowadays,biotherapy is one of effective treatments for cancer,which could enhance and adjust the body immunity and anti-cancer promotion ability through biological agent and method to inhibit and kill cancer cells.For the moment,cellular immunotherapy could not achieve idle clinical effect and still could not break through the bottleneck of treatment.What is more,in the process of adoptive cellular immunotherapy for cancer,although preferred DC-CIK cell could increase hundred times in vitro,it had unsatisfactory results.This case may be related to pay much attention to the expansion in vitro and ignore the immune activation in vivo.The main reason is that almost attention of research,at present,were forced on the activation of immunocompetent cell in last process of treatment,and ignoring the quality of immunocompetent cell and state of environment in vivo.Chinese medicine theory states that five organs producing each other and chi comes from Xiajiao but orig:inates from kidney.Based on this theory,we proposed a scientific hypothesis:the root of immune is not the self of immune but exterior of immune.If the quality of immune progenitor cells could be enhanced,the problem mentioned before would be solved.As the basic standpoint of tonifying kidney to enhance chi,clinical effects had been verify that prescription of Tonifying Kidney-Huiyang was superior to prescription of invigorating spleen and supplementing chi and others.The project preliminary studies shows that warm and tonify kidney-yang herbs could enhance the phagocytic immunity function.So based on these research results,we speculate that body immune function could be enhanced by stimulating of warm and tonify kidney-yang herbs.This study established model mouse of kidney-Yang deficiency induced by hydrocortisone and adopted prescription of Tonifying Kidney-Huiyang to active immune precursor cell of model mouses.We compared the change of associated lymphocyte,immune cell,lymphocyte subpopulation of normal mouses and model mouses with treated mouses through ELISA and FCM(flow cytometry)and other methods.At last,we systematically analyzed and researched these results and expect to find a new method to break through bottleneck of immunocyte therapy.This thesis consists of three parts.The first part describes the method of establishing model mouse of kidney-Yang deficiency and the effect of prescription of Tonifying Kidney-Huiyang on mice peripheral blood cytokines,including:IL-2,IL-4,IL-6,IL-10,INF-?,TNF and IL-17.The second part states the effect of prescription of Tonifying Kidney-Huiyang on mice peripheral blood T-lymphocyte(CD4+,CD8a+,CD4+/CD8a+)and NK cell(CD3-CD49b+).The third part includes the effect of prescription of Tonifying Kidney-Huiyang on mice associated lymphocyte.the main results are presented as follows:1.Prescription of Tonifying Kidney-Huiyang could protect model mouse of kidney-Yang deficiency induced by hydrocortisone through balancing the immunoregulation of Th1/Th2/Th17;2.Prescription of Tonifying Kidney-Huiyang could enhance the model mouse of kidney-Yang deficiency of the value of CD4+T lymphocyte,inhibit the expression of CD8+T lymphocyte and increase CD4+/CD8+;3.Prescription of Tonifying Kidney-Huiyang could decrease the model mouse of kidney-Yang deficiency of the ratio of neutrophile granulocyte and increase the ratio of lymphocyte.
