Objective:To observe the effect of point-moxibustion and electro-acupuncture on diabetic gastroparesis rats,explore the central regulation mechanism of two treatment methods for DGP,and to provide a new treatment for the prevention and treatment of DGP.Methods:40 SPF class SD rats were randomly allocated into blank group,model group,electro-acupuncture group and point-moxibustion group.DGP rat models were induced by high lipid and high glucose irregular breeding and intraperitoneal injection of streptozotocin(STZ)."Liangmen"(ST 21),"Zusanli"(ST 36),"Sanyinjiao"(SP 6)acu-points are selected among the electro-acupuncture group and point-moxibustion group.Information on the general physical signs scores,blood glucose,food intake after therapy was gathered and gastric emptying rate and small intestinal propulsion rate was measured.The relative protein expression and the mRNA expression of ghrelin,GHSR-1a were detected by Western blot and Rt-PCR.Results:1.General physical signs scores,blood glucose:Compared with the blank group,the model group significantly increased(p<0.01).Compared with the model group,the electro-acupuncture group and the point-moxibustion group reduced significantly(p<0.05);There was no significant difference between electro-acupuncture group and point-moxibustion group(p>0.05).2.Food intake,gastric emptying rate,small intestinal propulsion rate:Compared with the blank group,the model group decreased markedly(p<0.05,p<0.01);Compared with model group,electro-acupuncture group and point-moxibustion group increased obviously(p<0.05);and the curative effects of electro-acupuncture group and point-moxibustion group were inconspicuous(p>0.05).3.The relative protein expression and the mRNA expression of ghrelin,GHSR-1a:Compared with the blank group,the model group decreased markedly(p<0.05,p<0.01).The relative protein expression and the mRNA expression of ghrelin in point-moxibustion group were significantly higher than those in the model group(p<0.01),but there were no significant difference in GHSR-1a(p>0.05).Compared with that in the model group,the ghrelin and GHSR-1a in the electro-acupuncture group had no statistical significance(p>0.05).The relative protein expression and the mRNA expression of ghrelin in the point-moxibustion group were significantly different from those in the electro-acupuncture group(p<0.05).Conclusion:1.Electro-acupuncture and point-moxibustion can effectively relieve symptoms,reduce blood sugar,improve food intake and accelerate gastrointestinal motility.2.Activation of the hypothalamic arcuate nucleus-lateral septal nucleus ghrelin pathway by point-moxibustion can promote the expression of ghrelin thus improving gastric motility.
