Objective: This thesis is to discuss the therapeutic mechanism of abdominal acupuncture in treating gastroparesis syndrome caused by diabetes.The thesis is based on the experiment that studied the influence of GHRL and GHSR-la on lab rats falling in diabetic gastroparalysis(DGP).Methods: We randomly divided the lab rats into four different groups including the normal group,the model group,metoclopramide group and the needle group.DGP rat model was built on traperitoneal injection of streptozocin STZ in combination with high in fat and sugar irregular feeding.Then we observed the general indicators of lab rats after treatment,including measuring blood-sugar,Gastric emptying rate,intestinal propulsive rate,detecting GHSR-la ' s m RNA by means of RT-PCR,detecting GHSR-la's protein by western blot,and detecting the content of GHRL by EISA method.Results: 1.General symptoms: Compared with the blank group,the model group was thin,the fur was yellow and loose,the stool color became lighter,the stool traits became thinner,the smell was stinky,and the spirit was weak.Compared with the blank group,the symptom scores of the model group were increased(P<0.01);compared with the model group,the symptom scores of the metoclopramide group and the dull needle group were decreased(P<0.05 or P<0.01);There was no statistical difference between the dull needle group and the metoclopramide group(P>0.05).2.Blood sugar: Compared with the blank group,the blood sugar of the model group was increased(P<0.01).Compared with the model group,the blood sugar of the metoclopramide group and the dull needle group was decreased(P<0.01);There was no statistical difference between the dull needle group and the metoclopramide group(P>0.05).3.Gastric emptying rate and small intestine propulsion rate: Compared with the blank group,the gastric emptying rate and small intestine propulsion rate of the model group were decreased(P<0.01);Compared with the model group,the gastric emptying rate and small intestine propulsion rate of the metoclopramide group and the dull needle group were iecreased(P<0.05 or P<0.01).There was no statistical difference between the dull needle group and the metoclopramide group(P>0.05).4.GHRL,GHSR-1a protein expression and gene expression: Compared with the blank group,GHRL,GHSR-1a gene expression,protein expression of the model group were decreased(P<0.01);Compared with the model group,GHRL,GHSR-1a gene expression,protein expression of the metoclopramide group and the dull needle were increased by group(P<0.05 or P<0.01).There was no significant difference between the metoclopramide group and the dull needle group(P>0.05).Conclusion: 1.Dull Needle Method can improve the general symptoms,blood sugar and gastrointestinal motility of DGP rats.2.Dull Needle Method can promote the release of plasma GHRL and the expression of GHSR-1a protein and gene of the lateral septal nucleus,so that it can improve the gastrointestinal motility of DGP rats,which may be related to the activation of the arcuate nucleus of hypothalamic-Ghrelin pathway of lateral septal nucleus.
