Objective:To observe the effect of electro-acupunctrue(EA) ongeneral status score, blood sugar value, urine sugar value,gastro-intestinal hormone,such as serum insulin(INS),growth hormone (GH),cholecystokinin(CCK),Ghrelin, and ultrastructure of gastric antrum smooth muscle cells(GASMCs) in Diabetic Gastroparesis Model Rats, and to explore if can control blood sugar and improve gastric emptying in Diabetic Gastroparesis Model Rats with electro-acupuncture. What’s the relation of gastrointestinal hormone and the effects? To analysis the differences between the acupoints and non-acupoints, and to compare the acupoints specificity.Methods:forty-eight Sprague Dawley rats were randomly divided into4groups:normal contro(group A), DGP model(group B), EA point(group C), and EA non-point(group D), the number of every group is12rats. DGP rat model was established by2%STZ intraperitoneal injection at once, and then being fed with high-sugar-high-fat (HSHF) for8wks. using the OneTouch glucose meter and its test paper to measured the blood sugar, and using the urine sugar test paper to measured glycosuria, to note the general condition of rats score once a week. The rats were being treated with EA for15days. Phenol red lavage method was employed to measure gastric emptying rate (GER) and intestinal migration rate (IMR). Enzyme-linked immunoassay(Elisa) was employed to detect the level of insulin(INS) and growth hormone(GH), the content of cholecystokinin(CCK) and Ghrelin. The ultrastructure of GASMCs was observed under the electronic microscope.Results:1.The Diabetic Gastroparesis Model Rats were established successfully.Compared with the normal control group, the blood sugar and urine sugar in model group were increased significantly (P<0.01), the GER and IMR were decreased significantly(P<0.01). Compared with the model group, the blood sugar and urine sugar in EA points group were decreased significantly (P<0.01), the GER and IMR were increased significantly(P <0.01). Compared with the EA non-point group, the blood sugar and urine sugar in EA points group were not decreased significantly (P>0.05), the GER and IMR were increased significantly(P<0.05or P<0.01).2. the comparision of gastrointestinal hormone in each group rats: Compared with the normal control group, the level of serum insulin, growth hormone and the content of Ghrelin in model group were decreased significantly (P<0.05or P<0.01), the cholecystokinin were increased significantly(P<0.01). Compared with the model group, the level of serum insulin, growth hormone and the content of Ghrelin in EA points group were increased significantly (P<0.05or P<0.01), the cholecystokinin were decreased significantly(P<0.01). Compared with the EA non-points group, the level of serum insulin and the content of Ghrelin in EA points group were not increased significantly(P>0.05), the level of growth hormone was increased significantly(P<0.05). the cholecystokinin were decreased significantly(P<0.05).3. the comparision of ultrastructure of GASMCs in each group rats: Compared with the normal control group, the number of ICCs in model group were decreased significantly, the structure of smooth muscle cells were changed obviously. Compared with the model control group, the number of ICCs in EA points were increased as well as improving the ultrastructure of GASMCs.Conclusion:1.The urine sugar and blood sugar in model group were increased obviously, and the time of the GER and IMR were prolonged. Electro-acupuncture on "Zu San Li"(ST36) point, etc. has been decreased the sugar and promoted the movement of the gasrric antrum.2. Electro-acupuncture can improve the GI motility promisingly, based on a potential underlying mechanism that the electro-acupuncture can control the secretion of gastrointestinal hormone and improve the ultrastructure of gastric smooth muscle, and increase the number of ICCs.3.The effects of EA points were superior to the EA non-points group, it was said that the function of point is unique relatively.
