Objective:To observe effect of electroacupuncture(EA) at "Zusanli","Liangmen"(ST21),"Sanyinjiao"(SP6) on the general symptoms, sugar level, gastrointestinal motility and expression of Ghrelin and Growth Hormone Secretagogue Receptor(GHSR) mRNA in Diabetic Gastroparesis(Diabetic Gastroparesis,DGP)rats, so as to explore its mechanism underlying improvement of diabetic gastroparesis, and comparing the different between the effect of electroacupuncture and medicine. To provide an experimental basis for clinical application.Methods:DGP model rat was established by injection of55mg/kg,2%STZ into peritoneal cavity and raised by high-sugar and high-fat fodder irregularly for8weeks. Forty-eight SD rats were randomly and equally divided into normal control, model, EA Acupoint, metoclopramide groups,12rat/group.The general symptoms were assessed by scale, blood sugar was measured by glucometer, and the urine sugars were measured by Tes-Tape.After interventionthe rate of gastric emptying andpropelling rate of small intestine were measured by phenol red as markers, the expression of gastric antrum ghrelin and GHSR mRNA were examined by Real-time Quantitative PCR.Results:l.In comparison with the normal control group,the general symptoms of model group were general poor, blood sugar and urine sugar were markedly increased, the rate of gastric emptying, propelling rate of small intestine and the expression level of ghrelin mRNA and GHSR mRNA were decreased significantlyin model group (P<0.01,P<0.05).2.While in comparison with the model group, the general symptoms of EA acupoint group were improved, blood sugar and urine sugar were obviously decreased, the rate of gastric emptying, propelling rate of small intestine and the expression level of ghrelin mRNA and GHSR mRNA were increased markedly in the EA acupoint group (P<0.01,P<0.05).3.Compared with the model group, each index of the metoclopramide group were no significant different (P>0.05). Conclusion:1.Combination of55mg/kg2%STZ and high-sugar and high-fat irregular diets for8weeks could induce DGP rat model successfully.2.Electroacupuncture at ST36, ST21and SP6points can improve the general symptoms, blood sugar and urine sugar level of rats, promoted the gastrointestinal motility of DGPrats, which is closely associated with its effects in up-regulating the expression level of ghrelin mRNA and GHSR mRNA in gastric antrum.3.The effect of electroacupuncture at ST36, ST21and SP6points on promoting gastrointestinal motility in DGPrats was better than metoclopramide.
