Objective:To observe anti-inflammatory effect of moxibustion in the experimental RA rats and to explore the anti-inflammatory effect regulatory mechanism of mPD-1 and mPD-L1 for revealing the mechanism of moxibustion in the treatment of RA more depth.Methods:A total of 49 SD rats(200±20g weight,male and female each of half)were randomized blank-control group(blank group),RA model group(model group) and moxibustion treatment group(moxibustion group) with 15 cases in each.RA model was established by injection of Freund's Complete Adjuvant into the animal's bilateral joint cavities in the first day of the experiment.In the 7th day after building,moxibustion was applied to "Shenshu" (BL23) and "Zusanli" (ST36),5 cones every time, once daily (except Sundays),3 weeks altogether.Measuring the right foot swelling in the experiment first day(before building)and on the 7th,28th day.To detect the content of IL-10 and IFN-y in the peripheral blood serum with ELISA.Using the immunohistochemical method to detect the expression of mPD-1 and mPD-L1 in synovial tissue.Results:1.Moxibustion on "Shenshu"(BL23) and "Zusanli"(ST36) acupoints could bviously improve RA rats' right foot swelling. Synovial tissue pathology tests showed that:Compared with blank group,the hyperplasia and inflammatory cell infiltration of model group's synovial tissue and fibrous tissue were significantly increased (p<0.01).Compared with model group, moxibustion group were significantly lower (p<0.01).2.The test results of cytokines in peripheral blood serum show that:Compared with blank group,the content of IFN-y were remarkably higher (p<0.01) and IL-10 significantly decreased (p<0.01) in the model group.After moxibustion treating,the content of IFN-y was significantly lower (p<0.01) and IL-10 significantly increased (p<0.01) than mode group.3.The result of immunohistochemical showed that:Compared with blank group,model group's mPD-1 and mPD-L1 conent were significantly increased (p<0.05,p<0.01).The expression of mPD-1 and mPD-L1 were lower (p<0.01,p<0.05) than model group in moxibustion group.Conclusions:1.Moxibustion on"Shenshu"(BL23)and"Zusanli(ST36)"acupoints shows obvious anti-inflammatory effect in treating experimental RA.2.Moxibustion have a significant effect for RA rats IFN-? and IL-10 levels in serum,also affected the expression of mPD-1 and mPD-1 in synovial tissue. Moxibustion may adjust the expression of mPD-1 and mPD-L1 in synovial tissue, thereby inhibiting Pro-inflammatory cytokine IFN-y activity and promoting the secretion of anti-inflammatory factor IL-10, balancing Thl/Th2 cells, ultimately exert the anti-inflammatory effects.
