Anti-inflammatory Effect Of Treating Ra With Different Moxibustion Methods And Its Mechanism Of Heat Shock Protein 70 (hsp70)

Objective:On the basis of observing the different moxibustion methods(scarring moxibustion and moderate moxibustion) for experimental rheumatoid arthritis (RA) model rats'anti-inflammatory effect, to explore the status and function of HSP70 in the anti-inflammatory effects in different moxibustion methods。Methods:Fourty health SD rats (Sprague-Dawley rats) were randomly divided into control group (hereinafter referred to as "blank"), the RA model group (hereinafter referred to as "model"), moderate moxibustion treatment group (hereinafter referred to as " moderate moxibustion") and scarring moxibustion treatment group (hereinafter referred to as " scarring moxibustion"), each group of 10。To produce RA animal models by Freund's Complete Adjuvant (FCA), to moxibust in "Shenshu(BL23)" acupoints。Measuring foot swelling by foot volume, to detect IL-1,TNF-αcontent in the serum by enzyme-linked immunosorbent assay (ELISA), and HSP70 expression in the hypothalamus,inflammatory tissue by immunohistochemistry method.Results:(1) "Model", "moderate moxibustion" and "scarring moxibustion" right feet swelling increased significantly (P<0.01) compared with "blank" after modeling. After moxibustion treatment, right feet swelling of "moderate moxibustion" and "scarring moxibustion" were significantly lower than that of "model" (P<0.01), Compared with " moderate moxibustion ", " scarring moxibustion " right feet swelling had the markup tendency,but there were no significant difference (P>0.05). (2) Compared with "blank", "model" IL-1,TNF-αcontent were significantly increased (P<0.01). After moxibustion treatment, IL-1 and TNF-αcontent of "moderate moxibustion" and "scarring moxibustion" were significantly lower than that of "model" (P<0.01, P<0.05), Compared with " moderate moxibustion ", " scarring moxibustion " IL-1,TNF-αcontent in the serum had the markup tendency,but there were no significant difference(P>0.05). (3) Compared with "blank", "model" HSP70 expression in the hypothalamus were significantly lower (P<0.01), after moxibustion treatment, HSP70 expression HSP70 expression in the hypothalamus of "moderate moxibustion" and "scarring moxibustion" were significantly lower than that of "model" (P<0.01), but there were no significant difference between "moderate moxibustion" and "scarring moxibustion" (P>0.05); Compared with "blank", "model" HSP70 expression in the inflammatory tissue were significantly higher (P<0.01), after moxibustion treatment, HSP70 expression in the inflammatory tissue of "moderate moxibustion" and "scarring moxibustion" were significantly higher than that of "model" (P<0.01), but there were no significant difference between "moderate moxibustion" and "scarring moxibustion" (P>0.05).Conclusions:Different moxibustion (moderate moxibustion and scarring moxibustion) moxibust "Shenshu" acupoint on the experimental RA model rats'has significant anti-inflammatory effect, significantly decreased serum cytokines IL-1, TNF-αlevels, decreased HSP70 expression in the hypothalamus, increased HSP70 expression in inflammation tissue, but the anti-inflammatory effect of experimental RA model rats of different moxibustion (moderate moxibustion and scarring moxibustion) was no significant difference. Different moxibustion (moderate moxibustion and scarring moxibustion) produce anti-inflammatory effect may be due to thermal stimulation of moxibustion regulates the expression of HSP70 in the hypothalamus, which has stimulated a large number of HSP70 expression in inflammatory tissue that inhibited IL-1, tumor necrosis factor (TNF)-αand other inflammatory mediators expression.
