Objective:To observe the influence of Juanbi decoction on CTX-?,OC and COMP expression of the rabbit model of knee osteoarthritis, and explore the mechanisms in the treatment of rabbit osteoarthritis, then we can provide the theoretical basis for clinical treatment and further study.Methods:30 healthy Japanese white rabbits were divided into five groups at random:the control group(A),the model control group(B), the positive control group(C), the traditional medicine middle dose group(D) and the traditional medicine high dose group(E), 6 for each group. Group A was without operation. Other groups were duplicated into the rabbit model of knee osteoarthritis by Hulth's technique. All the rabbits moved with goad for 30 min everyday after 1 week. Group A and group B selected 2 rabbits model validated randomly 8 weeks after. If successful, the experiment was processing. Group A and group B were given equivalent normal saline, group C was given Diacerein Capsules,group D and group E were respectively given the Juanbi decoction with different concentrations. All drugs were given to the rabbits through stomach twice a day, curing for4 weeks continuously. Observing the changes of the rabbits in behavior and X ray and measuring the content of blood serum of CTX-?,OC and COMP during the experiment.Then the rabbits were sacrificed, Observing the pathological changes of arthroidal cartilage and being graded by Mankin's method and compared the differences between each two groups.Results : 1. Rabbits were without infection after the surgery, X-ray suggested duplicating successfully;2. Observing the rabbits' activity, the knees of rabbits in the group A were no deformation. When goaded the movements of the group A are normal. While knees of other groups rabbits were badly swollen, and arthroidal cartilages were deformation. When goaded, they were claudication in other groups.3. Observing with X-ray, effects in the groups C, D and E improved to varying degree after treatment compared with group B.4. General observation, the degeneration of the arthroidal cartilages improved to varying degree compared with group B, and the group E were improved significantly(P<0.01).Observing with microscope, the Mankin's scores of the group C and D were decreased compared with B, P<0.05. While the effect of group E was more significant, P<0.01.5. The contents of blood serum of CTX- ? and COMP in the group B were significantly increased compared with group A, P<0.01, while the OC content significantly decreased(P < 0.01).Group C and D were compared with group B, serum content of CTX-? and COMP were lower(P < 0.05), the OC content increased(P < 0.05). Compared with group B, group E CTX- ?, COMP content significantly decreased(P < 0.01), the OC content increased significantly(P < 0.01).Conclusion : 1.The modifie Hulth model technique can successfully establish the model of rabbit osteoarthritis. 2. Juanbi decoction can regulate the content of CTX-?,OC and COMP in blood serum effectively. 3. Through regulating the content of CTX-?,OC and COMP, Juanbi decoction can reduce the destruction of arthroidal cartilage, promote arthroidal cartilage repair, postpone the degeneration of arthroidal cartilage, relieve effectively clinical symptoms, then play an effect of prevention and treatment of OA.
