Objective: To study the effect of Dan Xuan Kou Kang drug on Oral Submucous Fibrosis and the expression of IL-17 A.Method: 1. 40 rats were divided randomly into 2groups. 8 normal rats is group A, the left is experimental group ? The rats of experimental group were established by injecting ANE into rats' oral submucouse for8 weeks to be the annimal model of oral submucous fibrosis. After that the experimental group divided randomly into 4 groups, group B, group C, group D, group E,8 rats in each. The rats in group B were killed directly to collect the Oral mucosal tissue samples.The other 3 groups respectively by gastric distilled water, low doses Danxuankoukang, high-dose Danxuankoukang for 4 weeks. During the experiment, rats' opening degree change were recorded. After drug intervention for 4 weeks, all rats were killed, and collected the Oral mucosal tissue samples to Observe histopathological changes and the expression of IL-17 A. Test the expression of hydroxyproline and IL-17 A.2. 30 rats were divided randomly into 3groups, 10 in each. Gavage physiological saline respectively, low doses Danxuankoukang, high-dose Danxuankoukang to get Preparation of normal and drug-containing serum. Cultivate oral mucosa fibroblasts of rats, use the fourth generation to do the study. Use normal serum, Danxuankoukang high and low doses of the serum and 200 ug/ml ANE to intervene for 48 h.. ELISA to detect the hydroxyproline content in the supernatant, and immunohistochemical method to detect the expression of IL-17 A.Result: Immunohistochemical SABC method to determine the expression of IL – 17 A found that Danxuankoukang groups can Reduce the expression of IL – 17 A,compared with the water group and the model group, there were significant differences(P<0.01); Hydroxyproline secretion in Dan Xuan Kou Kang drug-containing serum significantly lower than the ANE group(P<0.01); The expression of IL – 17 A in Danxuankoukang drug-containing serum group is significantly lower than the ANE group(P<0.01).Conclusion: The IL – 17 A might participate in the formation of OSF, Dan Xun Kou Kang can inhibit the expression of IL – 17 A to achieve the goal of treatment of OSF.
