Object:To investigate the qualitative and positioning expression of MMP-2protein in the buccal OSCC originating from OSF, and make sure that if the expression is related to its clinical features and relatively poor prognosis, and provide a theoretical basis for its targeted diagnosis and treatment.Materials&Methods:The experimental specimens were collected from the patients with buccal OSCC after radical surgical treatment (May2011-Feb2012) in the Department of Oral and Maxillofacial Surgery, Xiangya Hospital, Central South University. They were divided into3groups,19cases of buccal OSCC originating from OSF as the experimental group,16cases of buccal OSCC non-originating from OSF as a positive control group,15cases of normal buccal mucosa as a negative control group. The method of immunohistochemistry ABC was used to detect the qualitative and localized expression of MMP-2protein in the tissues of3groups, and the semi-quantitative was used to evaluate the expression level of MMP-2protein.Results:The expression of MMP-2protein was located in the cytoplasm and membrane of cancer cells, the IHC staining intensity of MMP-2protein in the cancer cells which located in the edge of cancer nest were stronger than the cells located in the central area of cancer nest.In the tissues of buccal OSCC originating from OSF, the expression of MMP-2protein were positive (the positive staining rate:94.97%); in the tissues of buccal OSCC non-originating from OSF was81.25%; the expression level and positive rate in the former group was slightly higher than the latter group, however, there was no statistical significance between the2groups(P>0.05).In the tissues of normal buccal mucosa, the expression of MMP-2protein were reduced or even disappeard(the positive rate:20%), there were statistically significant compared to the former2groups(P<0.05, P <0.05).There were no statistically significant between the following different clinical features such as cervical lymph node metastasis or not(P=0.886), clinical stage(stage Ⅰ Ⅱ and stage Ⅲ Ⅳ) according to the UICC(P=0.479), the size or primaty tumor(T1-T2and T3-T4)(P=0.379).Conclusions:1. The expression level of MMP-2was related to the invasion and metastasis of squamous cell carcinoma.2. The expression level of MMP-2protein in the tissues of buccal OSCC originating from OSF was higher than the tissues of buccal OSCC non-originating from OSF, it is consistent with its clinical and biological behavior of more invasive and metastatic, however, there was no statistically significant between the2groups, perhaps large sample for further studies is required.3. The expression level of MMP-2protein in the tissues of OSCC was higher than the tissues of normal buccal mucosa, the expression of MMP-2can be used as an indicator for clinical prognosis of patients with OSCC, and as a target for imaging diagnosis and treatment to guide clinical application.
