Ki-67 Expression In Oral Mucosal FB Cells Induced By ANE And Traditional Chinese Medicine Intervention

Objective: To observe the effect of serum containing Dan Xuan Kou Kang on oral mucous fibroblasts(FB)stimulated by areca nut extracts(ANE)proliferation and the expression of Ki-67 antigen.Methods: 1.30 SD rats were randomly divided into three groups,each group gavaged with physiological saline and DXKK(low dose?high dose)to make normal serum and drug-containing serum.2.Cultivate oral mucous fibroblasts,take the third or fourth generation cells used in the next experiment.1)oral mucous fibroblasts were determined by the method of SP.2)cells after inoculation were divided into 4 groups: A blank control group,B normal serum group,C medicated serum low dose group,D the drug-containing serum high dose groups,respectively added in normal serum,serum containing drugs and 100 ug/ml ANE,intervention in 48 hours.Determined by MTT to detect cell proliferation,record the absorbance value(OD);meantimes the expression of KI-67 in the cultured cells and the supernatant;Feulgen to detect DNA;Using microscopic to observe the micronucleus rate after Giemsa dyeing.Results: 1)the fibroblasts staining positive waveform can be seen in the cytoplasm protein.2)the average optical density value of FB in ANE stimulation is significantly higher than blank group,the drug-containing serum group OD value is lower than the normal group.3)KI-67 in FB and in supernatant fluid under ANE intervention is higher than that of blank group,group with drug-containing serum of KI-67 is lower than the normal serum group,the difference was significant(P<0.05).4)NA,NP,NV of FB in ANE stimulation is higher than the blank group(P<0.05),and the drug-containing serum group is lower than the normal serum group.5)the rate of micronucleus cells in FB cells under ANE intervention significantly higher than the blank group,Dan Xuan Kou Kang drug-containing serum group is lower than the normal serum group,and extent of the medicated serum high dose group is lower than the drug-containing serum of high dose group.Conclusion:1.100ug/ml ANE can promote the proliferation of FB cell.2.Dan Xuan Kou Kang drug-containing serum can affect the effect of ANE on proliferation of FB and high expression of KI-67.3.Dan Xuan Kou Kang can reduce Heteromorphism of FB under ANE and achieve the goal of treatment of OSF.
