Object: To observe the effect of Qi Wei Bai Zhu Powder on expression of IRF3、IRF7 and IFN in suckling mice infected by HRV.Method: 1. With a dilution of 10-7 HRV suspension, according to the literature [1] Oral infection 7-day-old suckling mice, 50μL/only. Normal suckling mice oral instillation of cell culture medium 50μL / only. Observations neonatal stool collected neonatal diarrhea stool by ELISA detection of fecal HRV, determine the success of modeling.2. Grouping and administration: diarrhea after suckling mice HRV infection model is successful, according to suckling mice nest randomly divided into model group(MG), Qiweibaizhu Powder Group(BG), ribavirin group(ZG), inhibitor group(YG), agonist group(JG), Qiweibaizhu powder plus inhibitor group(BYG), ribavirin plus inhibitor group(ZYG) and normal control group(NG)(8 groups). Each treatment group were suckling mice after oral instillation corresponding drug 50μL/(only. each time). treatment, the normal control group and model group orally dropped the same amount of cell culture medium3. The suckling mice were killed by neck breaking in 2h、8h、16h、24h、32h、48h after treatment and the content of IFN-a/b in serum were observed by ELISA.The positive expression of IRF3、IRF7、IFN-β in small intestinal mucosa epithelial cell were observed by using immunohistochemistry assay. CD4~+T、CD8~+T cell were separated from the small intestinal mucosa by immune magnetic bead(IMB).The expression of IRF3、IRF7 protein in CD4~+T、CD8~+T cell were observed by western blotting.Results: 1. Compared with NG, expression of IRF3、IRF7 in MG were increased at each time point(p<0.05); expression of IRF3、IRF7 in BG were significantly higher than those in MG、ZG(p<0.05).2. Compared with NG,expression of IRF3、IRF7 in MG were increased at each time point(p<0.05); expression of IRF3、IRF7 in BG were increased reached the climax at 24 h and significantly higher than those in MG、ZG(p<0.05).3. ELISA was used to detect the content of suckling mice serum IFN-α/β: the model group at each time point neonatal serum IFN-a /βwere higher than normal group(p<0.05); Qiweibaizhu powder group neonatal serum IFN-a content 32 h reached peak serum IFN-βcontent peaked at 24 h, with the model group and ribavirin comparison group, the difference was statistically significant(p <0.05); Qiweibaizhu powder plus inhibitor serum IFN-a /βcontent below Qiweibaizhu powder group, higher inhibitor group(p <0.05). By ELISA and immunohistochemistry to detect the expression of IFN-βsuckling mouse intestinal mucosa, and the result is consistent.Conclusion: Qiweibaizhu Powder through upregulation of suckling mice infected small intestine and small intestine lymphocytes IRF3, IRF7 expression,prompted an increase in serum IFN-a / β serum levels, and thus clear the HRV.
